大豆5個(gè)新發(fā)現(xiàn)ERF基因的生物信息學(xué)及表達(dá)分析

【摘要】:ERF轉(zhuǎn)錄因子廣泛存在于植物中并且參與植物對生物及非生物脅迫的響應(yīng)。從NCBI數(shù)據(jù)庫中獲得5個(gè)新的ERF基因,Gm ERFa/b/c/d/e。蛋白序列分析顯示,5個(gè)ERF基因均含有一個(gè)保守的AP2/ERF結(jié)合域。進(jìn)化分析表明,Gm ERFe與Gm ERF3和Gm ERF7同源性最高,Gm ERFb、Gm ERFc與Gm ERF5的同源性最高,Gm ERFd與Gm EREBP1的同源性較高,而Gm ERFa與其他ERF蛋白的同源性均較低。實(shí)時(shí)熒光定量PCR結(jié)果顯示,5個(gè)基因都主要在大豆的根和葉中表達(dá)。逆境處理后的實(shí)時(shí)熒光定量PCR結(jié)果顯示,Gm ERFd/e主要對乙烯信號和低溫產(chǎn)生響應(yīng),Gm ERFb主要對干旱產(chǎn)生響應(yīng),而Gm ERFa主要對低溫產(chǎn)生響應(yīng)。
[Abstract]:ERF transcription factors exist widely in plants and participate in plant response to biological and abiotic stress. Five new ERF genes were obtained from the NCBI database. Protein sequence analysis showed that all five ERF genes contained a conserved AP2/ERF binding domain. Evolutionary analysis showed that GM ERFe had the highest homology with GM ERF3 and GM ERF7. The homology between GM ERFd and GM ERF5 was higher than that between GM ERFd and GM EREBP1, but the homology between GM ERFa and other ERF proteins was lower. The results of real-time fluorescence quantitative PCR showed that the five genes were mainly expressed in the roots and leaves of soybean. The results of real-time fluorescence quantitative PCR showed that GM ERFd/e was mainly responsive to ethylene signal and low temperature. GM ERFb was mainly responsive to drought, while GM ERFa was mainly responsive to low temperature.
