甘藍(lán)型油菜PEPC基因ihpRNA表達(dá)載體的構(gòu)建與遺傳轉(zhuǎn)化研究

【摘要】:為提高油菜含油量,構(gòu)建了基于油菜磷酸烯醇式丙酮酸羧化酶基因(PEPC)的ihpRNA植物表達(dá)載體。分別從甘藍(lán)型油菜基因組中克隆了種子儲(chǔ)藏蛋白napin基因啟動(dòng)子(315 bp)、PEPC基因的外顯子片段(246 bp)及PEPC基因的內(nèi)含子(754 bp),包含了其剪切位點(diǎn)之前4 bp和之后18 bp片段。通過(guò)中間載體pBluescriptⅡSK(+)再構(gòu)建到pBI121中,構(gòu)成種子特異表達(dá)的內(nèi)含子剪接的PEPC基因的RNA干擾載體p BI121.NP+IP-。利用農(nóng)桿菌介導(dǎo)法對(duì)湘油15號(hào)進(jìn)行遺傳轉(zhuǎn)化,獲得卡那霉素抗性轉(zhuǎn)化植株9株,PCR檢測(cè)證實(shí)外源片段成功導(dǎo)入其中7株油菜基因組中。對(duì)T2種子中含油量檢測(cè)發(fā)現(xiàn)比對(duì)照提高10%左右。
[Abstract]:In order to improve the oil content of rape, the ihpRNA plant expression vector was constructed based on rape phosphoenolpyruvate carboxylase gene (PEPC). The exon of seed storage protein napin gene was cloned from the genome of Brassica napus, respectively, and the exon of PEPC gene was 754 BP, which contained 4 BP and 18 BP fragment before the cleavage site.
