大豆及豆制食品中轉(zhuǎn)基因成分檢測(cè)分析

【摘要】:建立大豆及豆制食品中轉(zhuǎn)基因成分分析的real-time PCR擴(kuò)增體系和方法。選擇市售的大豆、豆粉、豆干、豆腐、豆?jié){和腐竹等6種大豆及豆制食品,針對(duì)不同的樣品探索高質(zhì)量基因組DNA的提取方法,擴(kuò)增大豆內(nèi)參基因lectin進(jìn)行質(zhì)量檢測(cè)。針對(duì)我國批準(zhǔn)的3種主要轉(zhuǎn)基因大豆品系(GTS40-3-2,A2704-12和MON89788)設(shè)計(jì)特異性的引物和探針,進(jìn)行real-time PCR檢測(cè)。購買3種轉(zhuǎn)基因大豆品系的標(biāo)準(zhǔn)品作為質(zhì)控對(duì)照。從該6種大豆及豆制食品中均提取到了高質(zhì)量的基因組DNA,建立了穩(wěn)定的針對(duì)外源基因特異性、結(jié)構(gòu)特異性和品系特異性片段檢測(cè)的real-time PCR擴(kuò)增體系。成功建立了6種大豆及豆制食品中針對(duì)3種轉(zhuǎn)基因大豆品系成分檢測(cè)的real-time PCR方法。
[Abstract]:The real-time PCR amplification system and method for the analysis of transgenic components in soybean and soybean foods were established. Six kinds of soybean and soya bean foods, such as soybean, soybean powder, dried bean, bean curd, soybean milk and curd, were selected. The extraction methods of high quality genomic DNA were explored for different samples, and the lectin of soybean internal reference gene was amplified for quality detection. Specific primers and probes were designed for real-time PCR detection of three major transgenic soybean lines (GTS40-3-2A2704-12 and MON89788) approved in China. The standard samples of 3 transgenic soybean lines were purchased as quality control control. High quality genomic DNAs were extracted from the six soybean and soybean foods, and a stable real-time PCR amplification system was established for the detection of foreign gene specificity, structural specificity and strain specific fragments. A real-time PCR method for the detection of three transgenic soybean strains in 6 soybean and soybean foods was successfully established.
