小偃6號TaWRKY45基因在高溫抗條銹病中的功能研究

【摘要】:小偃6號是一個具有全生育期高溫抗條銹性的小麥品種。在小麥抗病研究中發(fā)現(xiàn)TaWRKY45轉(zhuǎn)錄因子參與了對多種病害的調(diào)控。為了研究TaWRKY45在小偃6號高溫抗條銹過程中的功能,以小偃6號為試驗材料,通過實(shí)時熒光定量PCR技術(shù)(Real-time quantitative PCR,qRT-PCR)研究TaWRKY45的表達(dá)模式,并利用病毒誘導(dǎo)的基因沉默(VIGS)技術(shù),研究了TaWRKY45沉默后對小偃6號高溫抗條銹病的影響。結(jié)果表明,TaWRKY45在小麥根、莖、葉中均有表達(dá),根中的表達(dá)量最高。當(dāng)小麥?zhǔn)艿剿畻钏?SA)、條銹菌、高溫(20℃)脅迫時,TaWRKY45表達(dá)量均上調(diào)。對小麥TaWRKY45基因進(jìn)行沉默處理后,沉默葉片(BSMV:TaWRKY45-as)的條銹菌菌落增長,壞死細(xì)胞數(shù)目減少。TaWRKY45正向參與了小偃6號的高溫抗條銹病過程。
[Abstract]:Xiaoyan 6 is a wheat variety with high temperature resistance to stripe rust in the whole growth period. In wheat disease resistance studies, TaWRKY45 transcription factors were found to be involved in the regulation of many diseases. In order to study the function of TaWRKY45 in the process of high temperature resistance to stripe rust of Xiaoyan 6, the expression pattern of TaWRKY45 was studied by real-time quantitative PCR technique (Real-time quantitative PCRQRT-PCRR), and the virus induced gene silencing (VIGS) technique was used. The effect of TaWRKY45 silencing on high temperature resistance to stripe rust of Xiaoyan 6 was studied. The results showed that TaWRKY45 was expressed in the roots, stems and leaves of wheat, and the highest expression was found in the roots.
