【摘要】：為研究LRR-RLKs亞家族基因RLK6(AT2G26730)在擬南芥生長(zhǎng)發(fā)育過(guò)程中的作用,本研究構(gòu)建了植物過(guò)表達(dá)載體p Super1300-RLK6,獲得5個(gè)過(guò)表達(dá)突變體,其中2個(gè)獨(dú)立株系(OE8,OE9)用于表型分析。通過(guò)對(duì)野生型(WT)、RLK6基因缺失突變體(rlk6-2、rlk6-3和rlk6-6)、過(guò)表達(dá)株系OE8和OE9生長(zhǎng)發(fā)育過(guò)程的詳細(xì)觀察和分析發(fā)現(xiàn),RLK6基因缺失突變體在整個(gè)生長(zhǎng)發(fā)育過(guò)程中的表型與WT差異不明顯,而過(guò)表達(dá)株系OE8和OE9比WT開(kāi)花早。此外,利用實(shí)時(shí)定量PCR(qRT-PCR)檢測(cè)各基因型植株中開(kāi)花相關(guān)基因的表達(dá)情況,結(jié)果表明,相對(duì)于WT,OE8和OE9中的開(kāi)花促進(jìn)基因CO、SOC1和LFY的表達(dá)量顯著升高,而開(kāi)花抑制基因FLC的表達(dá)量顯著下降。研究結(jié)果說(shuō)明RLK6過(guò)表達(dá)促進(jìn)了擬南芥的開(kāi)花,這為后續(xù)深入研究RLK6的作用機(jī)制奠定了基礎(chǔ)。
[Abstract]:In order to study the role of LRR-RLKs subfamily gene RLK6 (AT2G26730) in the growth and development of Arabidopsis thaliana, a plant overexpression vector, p Super1300-RLK6, was constructed, and five overexpression mutants were obtained, among which two independent lines were used for phenotypic analysis. Through the detailed observation and analysis of the growth and development process of the wild-type OE8 and OE9 lines, it was found that there was no significant difference between the phenotypic and WT phenotypes during the whole growth and development process of the wild-type OE8 / RLK6 gene deletion mutants, rlk6-2nrlk6-3 and rlk6-6, respectively. The overexpression lines OE8 and OE9 bloomed earlier than WT. In addition, the expression of flowering related genes in different genotypes was detected by real-time quantitative PCRQRT-PCR. the results showed that the expression of COOSOC1 and LFY in WTOOE8 and OE9 was significantly higher than that in WTOOE8 and OE9. However, the expression of flowering suppressor gene FLC decreased significantly. The results showed that the overexpression of RLK6 promoted the flowering of Arabidopsis thaliana, which laid a foundation for further study on the mechanism of RLK6.
