APEC感染雛雞小腸炎癥相關(guān)基因的RNA-Seq分析

【摘要】:【目的】研究禽致病性大腸桿菌(APEC)感染雛雞小腸炎癥相關(guān)基因表達(dá)的變化!痉椒ā繉(duì)14日齡雛雞腿部肌肉注射0.5mL/羽1×106 CFU/mL的APEC菌液,觀察雛雞的發(fā)病情況,并取病變嚴(yán)重雛雞空腸為測(cè)序樣品,同時(shí)以注射等量生理鹽水為對(duì)照,采用RNA高通量測(cè)序(RNA-Seq)技術(shù),篩選分析感染和未感染APEC雛雞小腸炎癥相關(guān)基因表達(dá)的變化。【結(jié)果】以差異倍數(shù)在2倍及以上為標(biāo)準(zhǔn),從試驗(yàn)組與對(duì)照組共篩選出差異表達(dá)基因131個(gè)。GO分類(lèi)結(jié)果顯示,有87個(gè)差異基因得到491個(gè)GO功能注釋,這些基因主要富集在炎癥反應(yīng)、肝素整合、白細(xì)胞介素-6的生物合成等過(guò)程;贙EGG分析,僅有29個(gè)差異基因得到注釋,涉及41個(gè)信號(hào)通路,主要參與的信號(hào)通路有PPAR、花生四烯酸代謝、MAPK等。【結(jié)論】APEC感染雛雞后會(huì)引起機(jī)體炎癥相關(guān)基因表達(dá)的變化,從中篩選出10個(gè)可能在炎癥反應(yīng)中具有重要影響的炎癥相關(guān)基因。
[Abstract]:[objective] to study the expression changes of inflammatory genes related to intestinal inflammation in chickens infected with avian pathogenic Escherichia coli (0.5mL/). [methods] to observe the incidence of 0.5mL/ plume (1 脳 10 6 CFU/mL) by intramuscular injection of 1 脳 10 6 CFU/mL APEC bacteria into the legs of 14 day old chicks. The jejunum of severely diseased chicks was taken as sequencing sample, and the same amount of saline was injected as control. The RNA high-throughput sequencing technique was used. The expression of inflammatory genes in small intestine of infected and uninfected APEC chicks was analyzed by screening and analyzing. [results] based on the standard of 2 times and more than 2 times of difference, 131 differentially expressed genes were selected from the experimental group and the control group. There are 87 differentially expressed genes with 491 go functional annotations. These genes are mainly enriched in inflammatory reaction, heparin integration, interleukin-6 biosynthesis and so on. Based on KEGG analysis, only 29 differentially expressed genes were annotated, involving 41 signaling pathways, mainly involved in PPAR, arachidonic acid metabolism and MAPK.Conclusion APEC infection may cause changes in the expression of inflammatory genes in chickens. Ten inflammatory genes that may play an important role in inflammatory response were screened.
