【摘要】：目的觀察claudin-6基因在結(jié)腸癌、結(jié)腸腺瘤及正常結(jié)腸黏膜組織中的表達(dá),以探討其與結(jié)腸癌發(fā)生發(fā)展的關(guān)系。方法應(yīng)用原位雜交法及免疫組織化學(xué)法對(duì)claudin-6 mRNA及其蛋白在結(jié)腸腺瘤(n=20)、結(jié)腸癌(n=42)及正常結(jié)腸組織(n=10)中的表達(dá)進(jìn)行檢測(cè)。結(jié)果 claudin-6 mRNA及其蛋白在結(jié)腸腺瘤及結(jié)腸癌組織中的陽(yáng)性表達(dá)率低于結(jié)腸正常黏膜,二者在結(jié)腸癌組織中的表達(dá)又低于結(jié)腸腺瘤,組間表達(dá)差異有統(tǒng)計(jì)學(xué)意義(χ~2=9.235,P0.05;χ~2=8.352,P0.05);claudin-6 mRNA及其蛋白的表達(dá)均與結(jié)腸癌組織的分化程度高低、有無(wú)淋巴結(jié)的轉(zhuǎn)移及浸潤(rùn)腸壁的深度有關(guān)(P0.05),二者在年齡、腫瘤大小及性別組內(nèi)的表達(dá)差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 claudin-6基因在結(jié)腸癌組織中呈低表達(dá),提示claudin-6表達(dá)的下調(diào)與結(jié)腸癌的發(fā)生、發(fā)展有密切關(guān)系。
[Abstract]:Objective to investigate the expression of claudin-6 gene in colon cancer, colon adenoma and normal colonic mucosa. Methods the expression of claudin-6 mRNA and its protein were detected by in situ hybridization and immunohistochemistry in adenoma of colon, adenoma nongma (42) and normal colonic tissue (10). Results the positive expression rate of claudin-6 mRNA and its protein in colon adenoma and colon cancer tissue was lower than that in colon normal mucosa, and the expression of claudin-6 mRNA and protein in colon cancer tissue was lower than that in colon adenoma. The expression of claudin-6 mRNA and its protein were correlated with the differentiation of colon cancer tissue, lymph node metastasis and the depth of invasion of intestinal wall (P 0.05). The expression of claudin-6 mRNA and its protein were related to the age of colonic carcinoma, and the expression of claudin-6 mRNA and its protein were related to the degree of differentiation, lymph node metastasis and the depth of invasion of intestinal wall. There was no significant difference in tumor size and expression in sex group (P 0.05). Conclusion the low expression of claudin-6 gene in colon cancer tissues suggests that the down-regulation of claudin-6 expression is closely related to the occurrence and development of colon cancer.
