豬流行性腹瀉病毒河南流行株S基因的克隆與遺傳進(jìn)化分析

【摘要】:近年來,我國大多省市暴發(fā)新生仔豬嚴(yán)重流行性腹瀉,給養(yǎng)豬業(yè)造成巨大經(jīng)濟(jì)損失。為分析豬流行性腹瀉病毒(PEDV)在河南省的流行和遺傳變異情況,本研究對(duì)2014—2015年收集的30份臨床腹瀉樣品,進(jìn)行RT-PCR檢測,對(duì)檢測為陽性的PEDV樣品進(jìn)行S基因的克隆與序列分析。結(jié)果表明,30份臨床腹瀉樣品中,RT-PCR檢測出22個(gè)PEDV陽性樣品,克隆的22條PEDV S序列與GenBank中發(fā)表的29條PEDVS基因序列核苷酸同源性為93.1%~99.2%。與經(jīng)典毒株相比,22株河南流行株在S1區(qū)域存在堿基的插入、缺失現(xiàn)象;S基因遺傳進(jìn)化樹表明,PEDV分為2群,河南流行株均屬于PEDVП群,與近年國內(nèi)分離株和韓國野毒株有較近親緣關(guān)系,而與歐洲株以及中國目前使用的疫苗株親緣關(guān)系較遠(yuǎn),基因差異較大。
[Abstract]:In recent years, severe epidemic diarrhea of newborn piglets broke out in most provinces and cities of China, which caused huge economic losses to pig industry. In order to analyze the prevalence and genetic variation of porcine epidemic diarrhea virus (PEDVV) in Henan Province, 30 clinical diarrhea samples collected from 2014-2015 were detected by RT-PCR, and the S gene was cloned and sequenced from the positive PEDV samples. The results showed that 22 PEDV positive samples were detected by RT-PCR in 30 clinical diarrhea samples. The nucleotide homology of 22 PEDV S sequences and 29 PEDVS gene sequences published in GenBank was 93.1% 99.2%. Compared with the classical strains, 22 Henan epidemic strains had base insertions in the S1 region. The genetic evolution tree of the deletion of the S gene indicated that the PEDV was divided into two groups, and all of the Henan epidemic strains belonged to the PEDV group. There is a close relationship with the domestic isolates and Korean wild strains, but with the European strains and the vaccine strains currently used in China, the genetic differences are relatively large.
