大豆GmAYI基因的克隆及初步功能預(yù)測(cè)

【摘要】:根據(jù)擬南芥At1G74730基因的保守序列設(shè)計(jì)引物,利用RT-PCR技術(shù)在大豆中克隆得到GmAYI基因。對(duì)GmAYI基因的結(jié)構(gòu)域和啟動(dòng)子元件進(jìn)行分析,結(jié)果表明:大豆GmAYI基因位于4號(hào)染色體上,含跨膜結(jié)構(gòu)域,屬于膜蛋白。該基因啟動(dòng)子區(qū)域含有激素相關(guān)應(yīng)答元件和胚乳表達(dá)所必須的順式作用元件。推測(cè)該基因可能參與調(diào)控大豆生長(zhǎng)發(fā)育及外界環(huán)境脅迫應(yīng)答等過(guò)程。進(jìn)而對(duì)14個(gè)大豆品種中GmAYI基因的轉(zhuǎn)錄水平進(jìn)行了定量分析,對(duì)GmAYI基因在14個(gè)大豆品種中的轉(zhuǎn)錄水平與單株粒數(shù)、單株粒重、百粒重、株高等產(chǎn)量相關(guān)性狀進(jìn)行相關(guān)性分析,結(jié)果表明:GmAYI基因的表達(dá)量與百粒重呈顯著負(fù)相關(guān),推測(cè)GmAYI可能是參與大豆產(chǎn)量相關(guān)性狀調(diào)控的基因。
[Abstract]:The GmAYI gene was cloned from soybean by RT - PCR . The results showed that the GmAYI gene could be involved in regulating the growth and development of soybean and the response of external environmental stress . The results showed that the expression of GmAYI gene was negatively correlated with the grain number , grain weight , grain weight , plant height , and so on .
