桑葉對糖尿病小鼠肝臟Toll樣受體基因表達的影響

【摘要】:目的:探討桑葉水提物對糖尿病小鼠肝組織Toll樣受體(Toll-like receptors,TLRs)基因表達量的影響,并探討其相關的作用機制。方法:ICR小鼠隨機分為正常組(6只),糖尿病組。糖尿病組小鼠利用高脂高糖飼料喂養(yǎng)10周聯合多次注射鏈脲佐菌素(STZ)建立高脂飼養(yǎng)和STZ共同誘導的小鼠糖尿病模型。造模成功小鼠隨機分為模型組,二甲雙胍組(600 mg·kg~(-1)),桑葉高、中、低劑量組(8,4,2 g·kg~(-1)),每組6只,灌胃(ig)給藥,每周測空腹血糖,于實驗中后期行口服糖耐量實驗,ig給藥10周后,測定小鼠血清胰島素、總膽固醇、甘油三酯和腫瘤壞死因子-α(TNF-α),并應用實時熒光定量PCR(Real-time PCR)檢測小鼠肝組織中TLR1~9表達量。結果:糖尿病模型小鼠經桑葉水提物治療后,進水量減少,顯著降低空腹血糖,改善糖耐量,顯著降低血脂,增加胰島素含量,改善胰島素抵抗,同時降低血清中TNF-α(P0.05,P0.01)。Realtime PCR結果顯示,糖尿病小鼠肝臟組織TLRs受體中TLR1,TLR2,TLR3,TLR7,TLR8和TLR9表達明顯高于正常組(P0.05,P0.01),TLR5表達明顯低于正常組;桑葉給藥組后,桑葉中劑量組肝組織中的TLR7,TLR8和TLR9表達量,顯著降低于模型組(P0.05,P0.01)。結論:桑葉水提物的降糖作用可能與抑制糖尿病模型小鼠肝組織中TLRs受體有關。
[Abstract]:Aim: to investigate the effect of mulberry leaf water extract on the expression of Toll-like receptor TLRs gene in liver tissue of diabetic mice and its related mechanism. Methods: ICR mice were randomly divided into normal group (n = 6) and diabetes group (n = 6). Diabetic mice were fed with high fat and high sugar diet for 10 weeks and were injected with streptozotocin (STZ) for 10 weeks to establish the diabetic model induced by hyperlipidemia and STZ. The successful mice were randomly divided into two groups: metformin group (600 mg / kg), mulberry leaf high, middle and low dose group (n = 6, n = 6, each group, n = 6). Fasting blood glucose was measured weekly, and oral glucose tolerance test was given 10 weeks later. Serum insulin, total cholesterol, triglyceride and tumor necrosis factor- 偽 TNF- 偽 were measured. The expression of TLR1~9 in liver tissue was measured by real-time fluorescence quantitative PCR(Real-time. Results: after treated with water extract from mulberry leaves, the intake of water decreased, fasting blood glucose decreased, glucose tolerance improved, blood lipids decreased, insulin content increased and insulin resistance improved. The expression of TLR7TLR8 and TLR7TLR8 in liver tissue of diabetic mice was significantly higher than that of normal group, and the expression of TLR7TLR8 and TLR9 in liver tissue of mulberry leaf medium dose group was significantly lower than that of normal control group, and the expression of TLR7TLR8 and TLR9 in liver tissue of mulberry leaf medium dose group was significantly lower than that of normal group after the administration of mulberry leaf, the expression of TLR7TLR8 and TLR9 in liver tissue of mulberry leaf treated group was significantly lower than that of normal control group, and the expression of TLR7TLR8 and TLR8 in liver tissue of mulberry leaf medium dose group was significantly lower than that of normal group. It was significantly lower than that of the model group (P 0.05, P 0.01). Conclusion: the hypoglycemic effect of mulberry leaf water extract may be related to the inhibition of TLRs receptor in liver tissue of diabetic mice.
