【摘要】：目的上皮-間質(zhì)轉(zhuǎn)化(EMT)是上皮來源的惡性腫瘤侵襲轉(zhuǎn)移的重要機制之一,其特征為上皮細胞標志物表達下調(diào)(E-cadherin等)而間質(zhì)細胞標志物(vimentin等)表達上調(diào)。本研究旨在探討人宮頸癌細胞系EMT相關(guān)標志物的表達情況,以確定宮頸癌細胞是否發(fā)生EMT,并為確定相關(guān)的細胞研究模型奠定基礎(chǔ)。方法采用逆轉(zhuǎn)錄PCR方法檢測4株人宮頸癌細胞系(SiHa、HeLa、RJC-1、CS1213)中E-cadherin、vimentin、Snail和Slug mRNA的表達;Western blotting檢測E-cadherin和vimentin的蛋白表達;免疫細胞化學(xué)方法檢測Snail和Slug的蛋白表達。結(jié)果vimentin和Snail mRNA在4種細胞中均有表達,E-cadherin和Slug mRNA在SiHa、HeLa和RJC-1細胞表達。E-cadherin蛋白在SiHa和RJC-1細胞表達,vimentin蛋白在HeLa和CS1213細胞表達。Snail蛋白在4個細胞株都有表達,Slug蛋白在SiHa、HeLa和RJC-1細胞表達。結(jié)論 HeLa和CS1213表現(xiàn)出間質(zhì)細胞表型,SiHa和RJC-1則表現(xiàn)為上皮細胞表型。
[Abstract]:Objective EMTs are one of the important mechanisms of invasion and metastasis of epithelial malignant tumors, characterized by down-regulated expression of epithelial cell markers (E-cadherin, etc.) and up-regulated expression of mesenchymal markers (vimentin). The purpose of this study was to investigate the expression of EMT related markers in human cervical cancer cell lines, and to determine whether or not EMTs occur in cervical cancer cells, and to lay a foundation for determining the relevant cell research models. Methods the expression of E-cadherin vimentinon Snail and Slug mRNA in four human cervical cancer cell lines, SiHaHN HeLaRJC-1rJC-1 and CS1213, was detected by reverse transcription PCR. The expression of E-cadherin and vimentin was detected by Western blotting, and the protein expression of Snail and Slug was detected by immunocytochemistry. Results the expression of E-cadherin and Slug mRNA in SiHa-HeLa and RJC-1 cells was found in both vimentin and Snail mRNA. E-cadherin protein was expressed in SiHa and RJC-1 cells. Snail protein was expressed in HeLa and CS1213 cells. Slug protein was expressed in SiHaHeLa and RJC-1 cells. Conclusion HeLa and CS1213 showed interstitial cell phenotype SIHA and RJC-1 as epithelial cell phenotypes.
