糯質(zhì)與非糯谷子Waxy基因exon4~exon9區(qū)間RT-PCR分析

【摘要】:為了明確糯質(zhì)和非糯谷子Waxy基因c DNA序列變異,為品質(zhì)育種提供實(shí)驗(yàn)依據(jù),以2份糯質(zhì)和2份非糯谷子為材料,利用特異RT-PCR和序列比對(duì),研究了Waxy基因exon4~exon9區(qū)間c DNA序列SNPs及其氨基酸多態(tài)性,結(jié)果表明,糯質(zhì)紅粘谷(00000537)與白米粘谷的同源性為99.12%,在第163、第192、第245和第358位點(diǎn)檢測(cè)到4個(gè)SNPs;非糯類(lèi)型豐田501與赤谷4號(hào)的同源性為98.9%,在第139、第189、第282、第445和第451位點(diǎn)有5個(gè)SNPs。糯質(zhì)紅粘谷(00000537)與非糯品種赤谷4號(hào)相似性為98.23%,紅粘谷(00000537)在第139、163、189、192、245、358、445和451位點(diǎn)堿基分別為T(mén)、G、T、G、G、A、C和A,赤谷4號(hào)在上述位點(diǎn)依次分別為C、A、G、A、A、C、T和G。糯質(zhì)紅粘谷(00000537)與非糯品種豐田501的相似性為98.9%,紅粘谷(00000537)在163、192、245、282和358位點(diǎn)分別為G、G、G、A和A,豐田501依次分別為A、A、A、G和C。紅粘谷(00000537)和赤谷4號(hào)exon4~exon9區(qū)間的c DNA序列編碼的氨基酸序列中,檢測(cè)出6個(gè)氨基酸變異,和豐田501出現(xiàn)3個(gè)氨基酸的變異。
[Abstract]:In order to identify the variation of c DNA sequence of Waxy gene in waxy and non-waxy grains, and to provide experimental basis for quality breeding, two waxy and two non-waxy grains were used as materials, and specific RT-PCR and sequence alignment were used. The polymorphism of SNPs and its amino acid in exon4~exon9 interval c DNA sequence of Waxy gene was studied. Four SNPs were detected at the 163th, 192nd, 245th and 358 sites, while the non-waxy type Toyota 501 had 98.9G homology with Chigu 4, and 5 SNPs at 139th, 189th, 282th, 445th and 45th. The similarity between glutinous red gluten and non-waxy cultivar Chigu 4 was 98.23, and the base pairs at site 139163189192245358445 and 451were respectively TGN and TGN, and Chigu No. 4 was CZAGAAACCYT and GG, respectively. The similarity between the glutinous red glutinous grain (00000537) and the non-waxy cultivar Toyota 501 was 98.9. The similarity between the glutinous rice and the non-waxy cultivar Toyota 501 was 163192245282 and 358, respectively. The similarity between the glutinous red glutinous grain and the non-waxy cultivar Toyota 501 was Geng Gan Gan A and A, and the Toyota 501 was Agna Agna and C respectively at the sites of 163192245282 and 358. Six amino acid variations were detected in the c DNA sequence in the exon4~exon9 region of Chigu 4, and 3 amino acids were detected in Toyota 501.
