氟化鈉對(duì)附睪形態(tài)結(jié)構(gòu)及緊密連接相關(guān)基因表達(dá)的影響

【摘要】:為了研究氟對(duì)小鼠附睪結(jié)構(gòu)形態(tài)及緊密連接相關(guān)基因的影響,將60只健康雄性小鼠隨機(jī)分成4組,對(duì)照組飲用蒸餾水,3個(gè)氟處理組分別飲用含氟化鈉(NaF)25、50和100mg/L的蒸餾水。染氟60d,取小鼠股骨,采用氟離子選擇電極法測(cè)其氟含量;取右側(cè)附睪制作石蠟切片,HE染色后進(jìn)行組織形態(tài)結(jié)構(gòu)觀察;左側(cè)附睪提取RNA,采用實(shí)時(shí)熒光定量PCR技術(shù)(qRT-PCR)檢測(cè)緊密連接相關(guān)基因的表達(dá)。結(jié)果顯示,與對(duì)照組相比,氟處理組小鼠股骨氟含量隨攝氟劑量的增加呈劑量依賴性,其中50、100mg/L劑量組差異極顯著(P0.01);氟處理組附睪尾管腔厚度極顯著增厚(P0.01);緊密連接相關(guān)基因Claudin-2 mRNA表達(dá)量極顯著下降(P0.01),100mg/L劑量組ZO-1mRNA表達(dá)量顯著下降(P0.05)。攝入過(guò)量NaF影響附睪形態(tài)及其緊密連接相關(guān)基因的表達(dá),從而可能對(duì)血-附睪屏障造成損傷。
[Abstract]:In order to study the influence of fluorine on the structure of epididymis and closely linked genes in mice, 60 healthy male mice were randomly divided into 4 groups. The control group drank distilled water and 3 fluoride treatment groups drank distilled water containing sodium fluoride (NaF) 25,50 and 100mg/L respectively. Fluorine 60d was used to take the femur bone of mice and the fluoride content was measured by fluorine ion selective electrode method. The paraffin section of the epididymis was made, and the tissue morphology and structure were observed after HE staining. The left epididymis was extracted with RNA, and the expression of closely linked genes was detected by real-time fluorescence quantitative PCR (qRT-PCR). The results showed that the fluoride content of the femur in the fluorine treatment group was dose-dependent with the increase of the dose of fluorine, of which 50100mg/L was compared with the control group. The difference in the dose group was very significant (P0.01); the thickness of the caudal caudal caudal lumen in the fluorine treatment group was significantly thicker (P0.01); the expression of Claudin-2 mRNA in the closely linked gene was significantly decreased (P0.01), and the ZO-1mRNA expression in the 100mg/L dose group decreased significantly (P0.05). The blood - epididymal barrier causes damage.
