【摘要】：目的探討尿路上皮癌乳腺癌易感基因1(BRCA1)mRNA水平與蛋白表達(dá)的相關(guān)性。方法收集50例尿路上皮癌患者標(biāo)本組織進(jìn)行石蠟切片3~5μm,采用免疫組織化學(xué)法檢測(cè)BRCA1基因蛋白的表達(dá)。通過(guò)提取石蠟切片組織RNA進(jìn)行反轉(zhuǎn)錄,采用實(shí)時(shí)定量PCR法檢測(cè)BRCA1基因mRNA的表達(dá)。結(jié)果 BRCA1 mRNA低表達(dá)率64%,中表達(dá)率36%;BRCAl免疫組織化學(xué)染色陽(yáng)性率為88%,陰性率為12%。采用SSPS 17.0統(tǒng)計(jì)學(xué)軟件,經(jīng)Wilcoxon兩樣本檢驗(yàn),BRCAl mRNA低表達(dá)組與中表達(dá)組的免疫組化染色程度差異有統(tǒng)計(jì)學(xué)意義(P0.000 1);Spearman等級(jí)相關(guān)分析顯示兩者呈正相關(guān)性(rs=0.652 9)。結(jié)論實(shí)時(shí)熒光定量PCR檢測(cè)mRNA表達(dá)與免疫組化檢測(cè)蛋白表達(dá)皆為評(píng)估尿路上皮癌的可選方法。
[Abstract]:Objective to investigate the correlation between 1(BRCA1)mRNA level and protein expression of breast cancer in urothelial carcinoma.Methods paraffin sections of 35 渭 m were collected from 50 patients with urothelial carcinoma and the expression of BRCA1 gene was detected by immunohistochemistry.The expression of BRCA1 gene mRNA was detected by real-time quantitative PCR method by extracting RNA from paraffin sections for reverse transcription.Results the low expression rate of BRCA1 mRNA was 64%, the middle expression rate was 36%. The positive rate of immunohistochemical staining of BRCAl was 88%, and the negative rate was 12%.Using SSPS 17.0 statistical software, two samples of Wilcoxon were used to test the difference in the degree of immunohistochemical staining between the low expression group of BRCAl mRNA and the middle expression group. There was a significant difference in the degree of immunohistochemical staining between the two groups. The correlation analysis showed that there was a positive correlation between the two groups.Conclusion Detection of mRNA expression by real-time fluorescence quantitative PCR and expression of protein by immunohistochemistry are the alternative methods for the evaluation of urothelial carcinoma.
