【摘要】：目的:探究糖尿病大鼠Nfic的表達(dá)狀況及其與其他成骨相關(guān)基因表達(dá)改變。方法:6周齡Wistar雄性大鼠30只,隨機(jī)分為對照組和糖尿病模型組。模型組大鼠腹腔注射鏈脲佐菌素(65mg/kg)建立1型糖尿病模型。觀察兩組大鼠體重、血糖變化;模型建成后8周處死大鼠,Q-PCR法檢測雙側(cè)股骨Nfic、Runx2、OSX(Oster ix)、COL2A1(Ⅰ型膠原)、OC(骨鈣素)、ALP、BMP-2、IGF-1(胰島素樣生長因子)的mRNA表達(dá)水平。結(jié)果:8周后,模型組大鼠體重降低或保持不變,對照組大鼠體重持續(xù)穩(wěn)定增長;模型組血糖持續(xù)保持在高血糖水平,對照組血糖位于正常范圍;Q-PCR檢測發(fā)現(xiàn),Nfic、Runx2、OSX、COL2A1、OC、IGF-1、BMP-2基因mR NA表達(dá)中,模型組明顯低于對照組。ALP基因的mRNA表達(dá)中,模型組顯著高于對照組(P0.01)。結(jié)論:1型糖尿病大鼠的高糖狀態(tài)使得Nfic等成骨相關(guān)基因表達(dá)下降,成骨標(biāo)記產(chǎn)物降低。
