陸地棉GST基因家族全基因組分析

【摘要】:谷胱甘肽轉(zhuǎn)移酶(glutathione-S-transferase,GST)是一種普遍存在的具有多功能的超家族蛋白,在植物初次生代謝、逆境脅迫、胞間信號(hào)傳遞等方面具有重要作用;同時(shí),作為配體其在植物激素代謝以及物質(zhì)轉(zhuǎn)運(yùn)方面也發(fā)揮作用。為了解析陸地棉(Gossypium hirsutum L.)GST基因家族的信息,本研究對(duì)該基因家族成員的種類、進(jìn)化關(guān)系、物理定位、基因結(jié)構(gòu)和保守基序以及表達(dá)模式進(jìn)行了分析。結(jié)果顯示,在陸地棉全基因組中共含有70個(gè)GST基因,進(jìn)化樹和基因結(jié)構(gòu)分析將該家族分為U族、F族、T族、Z族、EF1Bγ族和TCHQD族。基因定位分析發(fā)現(xiàn),除了AD/At2、AD/At4、AD/At5、AD/Dt5、AD/Dt10號(hào)染色體上沒有GST基因外,其他染色體上都有GST基因,并且在AD/At9、AD/Dt7、AD/Dt12、AD/Dt13這4條染色體上出現(xiàn)基因簇。對(duì)F族(Phi類)9個(gè)GST基因進(jìn)行熒光定量分析,結(jié)果表明,除Gh GSTF1可能為假基因外,Gh GSTF2~9等8個(gè)基因在陸地棉根、莖、葉以及各個(gè)發(fā)育時(shí)期的纖維中均有表達(dá);結(jié)合生物信息學(xué)分析,推測(cè)Gh GSTF8可能參與原花青素/花青素的轉(zhuǎn)運(yùn)和積累;Gh GSTF4、Gh GSTF6和Gh GSTF9可能在調(diào)節(jié)陸地棉的生長(zhǎng)和脅迫反應(yīng)中起作用,而Gh GSTF2、Gh GSTF3、Gh GSTF5和Gh GSTF7的功能還有待進(jìn)一步研究。本研究為陸地棉GST基因家族的分子進(jìn)化及功能研究提供了理論依據(jù)。
[Abstract]:Glutathione-S-transferase (GST) is a multifunctional superfamily protein, which plays an important role in plant primary metabolism, stress stress and intercellular signal transduction.As ligands, they also play a role in plant hormone metabolism and substance transport.In order to analyze the information of Gossypium hirsutum L.)GST gene family, the species, evolutionary relationship, physical location, gene structure, conserved motif and expression pattern of Gossypium hirsutum L.)GST gene family were analyzed in this study.The results showed that there were 70 GST genes in the whole genome of Upland cotton. The phylogenetic tree and gene structure analysis showed that the family was divided into U family, F family, T group, T family, group B, EF1B 緯 and TCHQD family.Gene mapping analysis showed that there was no GST gene on AD / At2 / AD / At4 / AD5 / Dt5 / AD5 / Dt5 / Dt10, and GST gene was found on all the other chromosomes, and there were gene clusters on the four chromosomes of AD / At9AD7 / Dt7 / AD12 / Dt13.The fluorescence quantitative analysis of 9 GST genes of Group F (Phi) showed that eight genes, including Gh GSTF2~9 and Gh GSTF2~9, were expressed in the roots, stems, leaves and fibers of Upland cotton with the possible pseudogenes of Gh GSTF1, and were analyzed by bioinformatics.It was suggested that Gh GSTF8 might be involved in the transport and accumulation of proanthocyanidins / anthocyanins. Gh GSTF6 and Gh GSTF9 might play an important role in regulating the growth and stress response of upland cotton, while the functions of Gh GSTF2 + Gh GSTF3Gh GSTF5 and Gh GSTF7 need to be further studied.This study provides a theoretical basis for the molecular evolution and functional study of GST gene family in upland cotton.
