【摘要】：目的克隆丹參Salvia miltiorrhiza中第14亞群R2R3 MYB轉(zhuǎn)錄因子基因SmMYB87的全長序列,并對其進(jìn)行生物信息學(xué)和表達(dá)分析。方法以丹參總RNA為模板,利用同源克隆和c DNA快速擴(kuò)增(RACE)技術(shù)獲得SmMYB87 cDNA全長序列。運(yùn)用生物信息學(xué)軟件對該基因及其編碼蛋白進(jìn)行結(jié)構(gòu)和理化性質(zhì)分析,利用q RT-PCR測定SmMYB87在各器官中的表達(dá)并構(gòu)建融合表達(dá)載體pTF-486-SmMYB87分析SmMYB87蛋白的亞細(xì)胞定位。結(jié)果 SmMYB87基因含有2個內(nèi)含子和1個732 bp的開放閱讀框(ORF),編碼243個氨基酸。該基因在根、莖、葉和花中均有表達(dá),且4個器官中的表達(dá)量沒有顯著差異。SmMYB87蛋白在細(xì)胞核和細(xì)胞膜上均有分布。結(jié)論 SmMYB87的序列結(jié)構(gòu)和表達(dá)分析為進(jìn)一步研究其在丹參中的生物學(xué)作用奠定了理論基礎(chǔ)。
[Abstract]:Objective to clone the full-length SmMYB87 gene of R2R3 MYB transcription factor gene from Salvia miltiorrhiza (Salvia) miltiorrhiza and analyze its expression and bioinformatics.Methods using total RNA of Salvia miltiorrhiza as template, the full-length sequence of SmMYB87 cDNA was obtained by homologous cloning and rapid amplification of C DNA.The structure and physicochemical properties of the gene and its encoded protein were analyzed by bioinformatics software. The expression of SmMYB87 in various organs was determined by Q RT-PCR and the fusion expression vector pTF-486-SmMYB87 was constructed to analyze the subcellular localization of SmMYB87 protein.Results the SmMYB87 gene contained 2 introns and 1 732 BP open reading frame, encoding 243 amino acids.The gene was expressed in root, stem, leaf and flower, and there was no significant difference in the expression of SmMYB87 protein in the nucleus and cell membrane.Conclusion the sequence structure and expression analysis of SmMYB87 lay a theoretical foundation for further study of its biological role in Salvia miltiorrhiza.
