ST釀酒酵母基因工程菌對大鼠腸道菌群的影響

【摘要】:為了研究ST釀酒酵母基因工程菌對大鼠腸道菌群的影響,試驗將90只SPF級SD大鼠隨機(jī)分為空白對照組、工程菌組及酵母菌組,分別灌服生理鹽水、工程菌菌液、釀酒酵母菌菌液,持續(xù)21 d后連續(xù)3 d灌服大腸桿菌H10407菌液,分別收集灌胃7,14,21天及攻毒后的各組大鼠糞便,提取糞便細(xì)菌DNA,利用末端限制性片段長度多態(tài)性(T-RFLP)、實時熒光定量PCR檢測了ST釀酒酵母基因工程菌對腸道菌群微生物可操作單元(OTU)和腸道5種重要細(xì)菌(類桿菌、梭菌、腸球菌、雙歧桿菌和乳桿菌)數(shù)量的變化,并測定了腸道菌群pH值。結(jié)果表明:在灌胃期間,工程菌組及酵母菌組的OTU值和雙歧桿菌、梭菌數(shù)量極顯著高于空白對照組(P0.01),其他細(xì)菌數(shù)量均不同程度升高。各組pH值均呈現(xiàn)下降趨勢,第21天時,工程菌組及酵母菌組pH值極顯著低于空白對照組(P0.01)。攻毒產(chǎn)腸毒素大腸桿菌后,工程菌OTU值和類桿菌、乳桿菌、雙歧桿菌數(shù)量均極顯著高于其他兩組(P0.01),pH值極顯著高于空白對照組(P0.01)。與灌服前相比,工程菌組的OTU值、細(xì)菌數(shù)量及pH值均變化幅度最小。說明ST釀酒酵母基因工程菌能夠改善腸道菌群結(jié)構(gòu),豐富腸道菌群多態(tài)性,降低pH值,增加有益菌群的數(shù)量。當(dāng)動物機(jī)體受到產(chǎn)腸毒素大腸桿菌感染時,ST釀酒酵母基因工程菌能夠有效降低產(chǎn)腸毒素大腸桿菌對腸道菌群的破壞,發(fā)揮預(yù)防保護(hù)作用,維持腸道內(nèi)菌群穩(wěn)定。
[Abstract]:In order to study the effect of genetically engineered strains of St Saccharomyces cerevisiae on intestinal flora of rats, 90 SD rats of SPF grade were randomly divided into three groups: blank control group, engineering bacteria group and yeast group.The liquid of Saccharomyces cerevisiae lasted 21 days and then was administered with Escherichia coli H10407 for 3 days. The feces of each group of rats were collected for 21 days after oral administration.Fecal bacterial DNA was extracted, and real-time fluorescent quantitative PCR was used to detect five important bacteria (Bacteroid bacillus, Clostridium spp, Enterococcus) and five important enteric bacteria (Bacteroid, Clostridium and Enterococcus), which were operated by ST-Saccharomyces cerevisiae.The number of bifidobacterium and Lactobacillus) and the pH value of intestinal flora were determined.The results showed that the OTU and Bifidobacterium counts of the engineering and yeast groups were significantly higher than those of the control group (P 0.01), and the other bacteria counts were increased in varying degrees during gastric perfusion.On the 21st day, the pH value of the engineering bacteria group and the yeast group was significantly lower than that of the blank control group (P 0.01).After enterotoxigenic Escherichia coli, the OTU value and the number of bacilli, Lactobacillus and Bifidobacterium were significantly higher than those of the other two groups.Compared with before administration, the OTU value, bacterial quantity and pH value of the engineering bacteria group were the smallest.The results showed that the genetically engineered strains of St Saccharomyces cerevisiae could improve the structure of intestinal flora, enrich the polymorphism of intestinal flora, reduce pH value and increase the number of beneficial flora.When the animal body is infected with enterotoxigenic Escherichia coli, the genetically engineered strain of S.cerevisiae St can effectively reduce the destruction of enterotoxigenic Escherichia coli to the intestinal flora, play a preventive and protective role, and maintain the stability of intestinal flora.
