【摘要】：微生物豐富多樣的次級(jí)代謝產(chǎn)物一直都是天然藥物的重要來(lái)源。隨著微生物基因組學(xué)研究的深入,人們發(fā)現(xiàn)在現(xiàn)有的培養(yǎng)條件下很多生物合成基因簇未能表達(dá),從而無(wú)法生成相應(yīng)的代謝產(chǎn)物。這些處于沉默狀態(tài)的基因簇給新型藥物的開(kāi)發(fā)帶來(lái)了新的契機(jī)。本文綜述了激活這些沉默基因簇的三種主要方法:調(diào)控基因改造、強(qiáng)啟動(dòng)子引入及小分子物質(zhì)添加。激活微生物中沉默基因簇將有望得到結(jié)構(gòu)新穎、活性顯著的新活性分子。
[Abstract]:Microbial secondary metabolites are always important sources of natural drugs.With the development of microbial genomics, it has been found that many biosynthetic gene clusters can not be expressed under the existing culture conditions, so they can not produce the corresponding metabolites.These silent gene clusters offer new opportunities for the development of new drugs.This paper reviews three main methods for activating these silencing gene clusters: regulation of gene transformation, introduction of strong promoters and addition of small molecules.Activation of silencing gene clusters in microbes is expected to lead to new active molecules with novel structure and significant activity.
