【摘要】：旨在研究雙鏈RNA(Double-stranded RNA,dsRNA)對(duì)海蘭褐蛋雞卵泡可卡因-苯丙胺調(diào)節(jié)轉(zhuǎn)錄肽(Cocaine-and amphetamine-regulated transcript,CART)基因表達(dá)及雌激素(Estrodiol,E2)和孕酮(Progestin,P)分泌的影響。選擇60只20周齡海蘭褐蛋雞,全價(jià)日糧預(yù)飼30d后,隨機(jī)平均分為3個(gè)處理組:對(duì)照組(A組)、dsRNA每5d注射1次(B組)、dsRNA僅第1天注射1次(C組);30d后每組各選擇10只健康蛋雞宰殺,分別采集3~5、6~8、9~12mm卵泡,提取總RNA進(jìn)行qRT-PCR檢測(cè);并對(duì)E2和P濃度進(jìn)行測(cè)定。結(jié)果顯示:注射dsRNA后,處理組B和C組CART表達(dá)量與對(duì)照組相比均降低,且在6~8 mm卵泡CART表達(dá)量顯著低于對(duì)照組(P0.05);B組注射dsRNA后,可顯著提高6~8、9~12mm卵泡E2的分泌(P0.05);但各處理組對(duì)各卵泡P濃度無(wú)顯著影響。表明體外靜脈注射dsRNA可抑制蛋雞卵泡CART的表達(dá),并進(jìn)一步通過促進(jìn)卵泡細(xì)胞E2的分泌來(lái)影響卵泡的發(fā)育。
[Abstract]:Sixty 20-week-old Hailan brown laying hens were randomly divided into three treatment groups after 30 days of prefeeding the whole ration: control group A was treated with dsRNA once every 5 days and group B was treated with dsRNA once every 5 days. After 30 days of treatment, 10 healthy laying hens were slaughtered in each group.The follicles were collected from the follicles of 3 pieces and 5 pieces, and the total RNA was extracted for qRT-PCR detection, and the concentrations of E 2 and P were determined.The results showed that the expression of CART in group B and C was significantly lower than that in group B and C after injection of dsRNA, and the expression of CART in 6mm follicle was significantly lower than that in group B (P 0.05).The E _ 2 secretion of 12 mm follicles increased significantly (P 0.05), but there was no significant effect on the P concentration of each follicle in each treatment group.The results showed that dsRNA could inhibit the expression of CART and further affect the development of follicles by promoting the secretion of E2 in follicle cells.
