【摘要】：為分析牛乳頭狀瘤病毒2型貴州株(BPV2-GZ01株)L2基因的分子特征,預(yù)測(cè)編碼蛋白的生物學(xué)功能,對(duì)BPV2-GZ01株L2基因進(jìn)行PCR擴(kuò)增、克隆及序列測(cè)定,應(yīng)用生物信息學(xué)相關(guān)軟件及方法,對(duì)BPV2-GZ01株L2基因進(jìn)行序列分析并對(duì)其編碼蛋白進(jìn)行二級(jí)結(jié)構(gòu)、三級(jí)結(jié)構(gòu)、B細(xì)胞表位、保守結(jié)構(gòu)域、跨膜結(jié)構(gòu)域和信號(hào)肽預(yù)測(cè)。結(jié)果顯示,L2基因PCR擴(kuò)增產(chǎn)物大小為1404 bp,編碼467個(gè)氨基酸;與參考株BPV2株、BPV2-SW01株、BPV2-AKS01株、BPV13株、BPV1株相應(yīng)序列核苷酸同源性分別為99.0%、99.7%、99.3%、83.9%和75.1%,氨基酸的同源性分別為98.9%、99.6%、99.4%、89.5%和82.7%;系統(tǒng)進(jìn)化顯示,BPV2-GZ01株L2基因與BPV2-SW01株親緣關(guān)系最近;二級(jí)結(jié)構(gòu)以無規(guī)則卷曲、β-折疊和α-螺旋區(qū)域所占比例較大,預(yù)測(cè)此蛋白可能存在B細(xì)胞優(yōu)勢(shì)抗原表位,無跨膜區(qū)域,無信號(hào)肽區(qū)域。本研究結(jié)果將為貴州省BPV核酸疫苗研究提供理論依據(jù)。
[Abstract]:In order to analyze the molecular characteristics of BPV2-GZ01 strain of bovine papillomavirus type 2 (BPV2-GZ01) and predict the biological function of encoding protein, the L2 gene of BPV2-GZ01 strain was amplified by PCR, cloned and sequenced, and the relevant software and methods of bioinformatics were used.The L2 gene of BPV2-GZ01 strain was sequenced and its encoding protein was predicted by secondary structure, tertiary structure, B cell epitope, conserved domain, transmembrane domain and signal peptide.The results showed that the PCR amplification product of L2 gene was 1404 BP, encoding 467 amino acids.The secondary structure was characterized by irregular curl, 尾 -folding and 偽 -helix. It was predicted that the protein might have B cell epitopes, no transmembrane region and no signal peptide region.The results of this study will provide theoretical basis for the study of BPV nucleic acid vaccine in Guizhou province.
