【摘要】：兩性霉素B(AmB)作為第一個用于深部真菌感染的藥物,自上市以來已使用超過半個世紀,目前仍是臨床治療中不可缺少的藥物。隨著近年來AmB衍生物藥物的發(fā)展,以及其生產菌株結節(jié)鏈霉菌(Streptomyces nodosus)全基因組和AmB體內合成代謝途徑的解析,運用基因工程和代謝工程的方法對AmB組合生物合成的研究逐漸增多。由于AmB在當今臨床治療上的地位仍極其重要,因此促進AmB產業(yè)的發(fā)展具有顯著的經(jīng)濟及社會效益。本文對AmB進行了概述,并對其合成基因簇相關基因及其當前組合生物合成的研究現(xiàn)況進行了總結與探討。
[Abstract]:Amphotericin (Amb), the first drug used for deep fungal infection, has been used for more than half a century since it came into the market, and is still an indispensable drug in clinical treatment.With the development of AmB derivative drugs in recent years, and the analysis of the whole genome of Streptomyces nodosus, the biosynthesis of AmB by genetic engineering and metabolic engineering methods, the biosynthesis of AmB has been gradually increased.As the status of AmB is still very important in current clinical treatment, it has significant economic and social benefits to promote the development of AmB industry.In this paper, AmB is reviewed, and the research status of its gene cluster related genes and its combination biosynthesis is summarized and discussed.
