TNFRSF1A基因CpG島甲基化與基因表達(dá)及T淋巴細(xì)胞亞群指標(biāo)的相關(guān)性分析

【摘要】:旨在探討TNFRSF1A基因CpG島的甲基化水平,闡明該基因甲基化與基因表達(dá)及T淋巴細(xì)胞亞群性狀的關(guān)系。本研究以抗病力強的華北型黑豬大蒲蓮豬和抗病力相對較弱的引進(jìn)豬種長白豬的仔豬為試驗群體,通過亞硫酸氫鹽處理后測序法(BSP)探求TNFRSF1A基因上游CpG島甲基化情況,并將甲基化位點的甲基化頻率與基因表達(dá)量及T淋巴細(xì)胞亞群性狀進(jìn)行相關(guān)性分析。結(jié)果表明:1)TNFRSF1A基因起始位點附近預(yù)測到2個CpG島(Island),跨這2個CpG島進(jìn)行BSP測序后共發(fā)現(xiàn)25個CpG位點,其中Island 1和Island 2中各有9個CpG位點,其余位于2個CpG島之間。Island 1的甲基化頻率在大蒲蓮和長白豬兩群體間差異不顯著(P0.05),Island 2的甲基化頻率大蒲蓮豬群體顯著高于長白豬群體(P0.05)。2)Island 2中甲基化頻率與ΔCt值成正相關(guān),即甲基化頻率與基因表達(dá)量成負(fù)相關(guān),其中+126、+128、+159、+162和+164是重要的甲基化位點。3)兩個豬群體中位于Island 2的+159和+164位點與CD4+CD8+CD3+指標(biāo)顯著正相關(guān)(P0.05)。Island 2中+159和+164位點甲基化頻率與基因表達(dá)量成負(fù)相關(guān),而且這2個位點又與T淋巴細(xì)胞亞群CD4+CD8+CD3+顯著正相關(guān),筆者推測,大蒲蓮仔豬TNFRSF1A基因+159和+164位點的甲基化頻率增加,可能導(dǎo)致TNFRSF1A基因表達(dá)量下降,影響T淋巴細(xì)胞亞群性狀,從而影響機(jī)體的免疫水平。
[Abstract]:The aim of this study was to investigate the methylation level of TNFRSF1A gene CpG island and to elucidate the relationship between methylation and gene expression and T lymphocyte subsets.In this study, we studied the methylation of CpG islands in the upper reaches of TNFRSF1A gene by using high-resistant North China black pig Pu Lin and piglets with relatively weak resistance to disease of imported Landrace pigs.The correlation between methylation frequency and gene expression and T lymphocyte subsets was analyzed.The results showed that two CpG islands were predicted near the initiation site of TNFRSF1A gene, and 25 CpG loci were identified by BSP sequencing across the two CpG islands, including 9 CpG loci in Island 1 and Island 2, respectively.There was no significant difference in methylation frequency between Dapulian and Landrace pigs in the other two CpG islands. The methylation frequency of Dapu Island 2 was significantly higher than that of Landrace Pig population (P 0.05). The methylation frequency was positively correlated with 螖 Ct value in Landrace White Pig population (P0.05) and Landrace Island 2, respectively.That is, the frequency of methylation is negatively correlated with the amount of gene expression.Among them, 126, 128, 159, 162 and 164 were important methylation sites. 3) there was a negative correlation between the methylation frequencies of 159 and 164 loci in Island 2 and the CD4 CD8 CD3 index.Moreover, these two loci were positively correlated with CD4 CD8 CD3 of T lymphocyte subsets. We speculate that the increase of methylation frequency of TNFRSF1A gene 159 and 164 in Pu Lin piglets may lead to the decrease of TNFRSF1A gene expression.The effect of T lymphocyte subsets, thus affecting the immune level of the body.
