水稻干尖線蟲海藻糖酶Ab-tre-1基因克隆與逆境條件下的表達(dá)分析

【摘要】:海藻糖酶參與生物體內(nèi)能量代謝,在抵御逆境脅迫過程中發(fā)揮關(guān)鍵作用。水稻干尖線蟲是水稻上重要的寄生線蟲,能夠在多種逆境中存活,嚴(yán)重危害水稻生長(zhǎng)。為探究水稻干尖線蟲海藻糖酶基因的功能,利用RACE-PCR技術(shù)獲得了該線蟲海藻糖酶基因全長(zhǎng),將其命名為Ab-tre-1,并利用實(shí)時(shí)熒光定量PCR技術(shù)分析在線蟲卵、不同齡期和若干逆境條件下該基因的表達(dá)情況。結(jié)果表明,該基因DNA序列長(zhǎng)2 083bp,包含9個(gè)內(nèi)含子,開放閱讀框1 743 bp,編碼580個(gè)氨基酸,含有2個(gè)典型的海藻糖酶標(biāo)簽基序。進(jìn)化樹結(jié)果表明,水稻干尖線蟲海藻糖酶與自由生活線蟲和動(dòng)物寄生線蟲位于不同進(jìn)化分支。Ab-tre-1在線蟲卵中的表達(dá)最高,二齡幼蟲和成蟲次之,三、四齡幼蟲表達(dá)水平較低。此外,當(dāng)水稻干尖線蟲暴露于干燥、活性氧、高溫和藥劑脅迫環(huán)境中,Ab-tre-1表達(dá)水平均顯著提高,表明該基因可能與水稻干尖線蟲的抗逆過程有關(guān)。本研究結(jié)果為闡明水稻干尖線蟲海藻糖酶基因抗逆表達(dá)提供了理論依據(jù)。
[Abstract]:Trehalase is involved in energy metabolism and plays a key role in resisting stress.Rice dry tip nematode is an important parasitic nematode in rice. It can survive many kinds of stresses and seriously harm the growth of rice.In order to study the function of trehalase gene of rice dry tip nematode, the full length of trehalase gene was obtained by RACE-PCR technique, named Ab-tre-1, and real-time fluorescence quantitative PCR was used to analyze the eggs.The expression of the gene in different ages and some stress conditions.The results showed that the DNA sequence of the gene was 208.3 BP, containing 9 introns, open reading frame 1,743 BP, encoding 580amino acids, and containing two typical trehalase tag motifs.The results of phylogenetic tree showed that the expression of trehalase, free living nematodes and parasitic nematodes were the highest in the eggs of different evolutionary branches. The second instar larvae and adults were the second, and the third and fourth instar larvae had lower expression level.In addition, the expression level of Ab-tre-1 was significantly increased in dry, reactive oxygen species, high temperature and pesticide stress environments, suggesting that the gene might be related to the stress resistance process of rice dry tip nematode.The results of this study provide a theoretical basis for clarifying the stress resistance of trehalase gene of rice dry tip nematode.
