【摘要】:目的:研究KIF20A對肝癌SMMC-7721細胞系增殖和侵襲遷移能力的影響。方法:利用RNA干擾技術(shù)下調(diào)肝癌細胞系SMCC-7721中KIF20A的表達。qRT-PCR和Western blot分別檢測細胞KIF20A在mRNA和蛋白水平表達中的變化。CCK-8實驗檢測增殖能力的改變。Transwell實驗檢測遷移、侵襲能力的改變。結(jié)果:qRT-PCR和Western blot結(jié)果顯示KIF20A-siRNA能夠成功下調(diào)KIF20A的表達,CCK-8和Transwell實驗分別證實肝癌SMMC-7721細胞增殖和侵襲遷移能力下降。結(jié)論:抑制KIF20A的表達能夠降低肝癌細胞的增殖和侵襲遷移能力。
[Abstract]:Objective: to study the effect of KIF20A on the proliferation, invasion and migration of hepatocellular carcinoma SMMC-7721 cell line.Methods: the expression of KIF20A in hepatocellular carcinoma cell line SMCC-7721 was down-regulated by RNA interference technique. QRT-PCR and Western blot were used to detect the changes of KIF20A expression at mRNA and protein levels respectively. CCK-8 assay was used to detect the change of proliferation ability. Transwell assay was used to detect migration and invasion ability.Results the results of KIF20A-siRNA and Western blot showed that KIF20A-siRNA could successfully down-regulate the expression of KIF20A. CCK-8 and Transwell experiments confirmed that the proliferation, invasion and migration of SMMC-7721 cells decreased respectively.Conclusion: inhibiting the expression of KIF20A can reduce the proliferation, invasion and migration of hepatoma cells.
