【摘要】：通過(guò)海島棉基因組數(shù)據(jù)庫(kù)和GSDS 2.0在線生物信息學(xué)軟件,分析GbHCT基因的結(jié)構(gòu),利用實(shí)時(shí)熒光定量PCR技術(shù),檢測(cè)GbHCT基因在海島棉不同組織和棉纖維不同發(fā)育時(shí)間的表達(dá)量。結(jié)果表明,該基因DNA序列長(zhǎng)度為1 953bp,開(kāi)放閱讀框長(zhǎng)度為1 311bp,編碼436個(gè)氨基酸,在海島棉基因組中對(duì)應(yīng)scaffold3453序列,含有1個(gè)內(nèi)含子和2個(gè)外顯子。GbHCT基因在不同組織和纖維發(fā)育不同時(shí)期中均有表達(dá),在各個(gè)組織中,苞葉和花中表達(dá)量最高,在棉纖維不同發(fā)育時(shí)期開(kāi)花后5d和25d表達(dá)量最強(qiáng)。該基因可能參與棉纖維發(fā)育伸長(zhǎng)期和次生壁增厚期。為深入研究該基因的功能,構(gòu)建植物表達(dá)載體pEGAD-GbHCT并轉(zhuǎn)入根癌農(nóng)桿菌,為下一步研究奠定試驗(yàn)基礎(chǔ)。
[Abstract]:The structure of GbHCT gene was analyzed by genomic database of island cotton and GSDS 2.0 online bioinformatics software. The expression of GbHCT gene in different tissues and fiber of island cotton was detected by real-time fluorescence quantitative PCR technique.The results showed that the length of the DNA sequence was 1 953 BP and the open reading frame length was 1 311 BP, encoding 436 amino acids, corresponding to the scaffold3453 sequence in the genome of sea island cotton.GbHCT gene with one intron and two exons was expressed in different tissues and at different stages of fiber development. The highest expression was found in bracts and flowers, and the highest in 5 and 25 days after flowering in different development stages of cotton fibers.This gene may be involved in cotton fiber extension and secondary wall thickening.In order to further study the function of the gene, the plant expression vector pEGAD-GbHCT was constructed and transferred into Agrobacterium tumefaciens.
