未控制哮喘與ADRB2基因位點多態(tài)性關(guān)系的分析

【摘要】:目的:探討β2腎上腺素能受體(β2-adrenergicreceptor,ADRB2)的基因rs1042718、rs2895795及rs12654778位點基因多態(tài)性與未控制哮喘的相關(guān)性。方法:選取2015年12月到2016年12月我院確診為支氣管哮規(guī)范化治療半年后未控制者30例為病例組,完全控制及部分控制者30例為陰性對照組,同期我院健康體檢者30例為正常對照組,采用Sequenom MassARRAY?SNP檢測法和基質(zhì)輔助激光解吸附電離飛行時間質(zhì)譜分析質(zhì)譜技術(shù)對三個位點進(jìn)行多態(tài)性檢測。結(jié)果:rs12654778位點存在GG/GA/AA三種基因型,其基因型的分布頻率在三組無統(tǒng)計學(xué)意義(GG/GA/AA)且其基因型及等位基因的分布頻率在三組間比較差異無統(tǒng)計學(xué)意義(P=0.554/0.297/0.819);rs2895795位點TT/AT/AA三種基因型,在三組間比較差異無統(tǒng)計學(xué)意義(P=0.105/0.104/0.730);rs1042718位點基因型三組間比較差異無統(tǒng)計學(xué)意義(P=0.141/0.051/0.846)。結(jié)論:ADRB2基因多態(tài)性與未控制哮喘發(fā)病無明顯相關(guān)性,這可能與種族差異、單倍型有關(guān)。
[Abstract]:Objective: To investigate the effect of beta 2 adrenergic receptor (beta 2-adrenergicreceptor, ADRB2) genes rs1042718, rs2895795 and the correlation between rs12654778 gene polymorphism and uncontrolled asthma. Methods: from December 2015 to December 2016 in our hospital diagnosed as bronchial asthma standardized treatment after six months without control in 30 cases as the case group, completely control and control part 30 cases were negative control group in our hospital during the same period, 30 healthy subjects served as normal control group, using Sequenom MassARRAY? SNP detection method and matrix assisted laser desorption ionization time-of-flight mass spectrometry mass spectrometry analysis was used to detect the polymorphism of three loci. Results: rs12654778 loci showed GG/GA/AA three genotypes and its genotype the frequency distribution in the three groups was not statistically significant (GG/GA/AA) had no statistical significance and the frequency distribution of genotypes and allele comparison in the differences between the three groups (P=0.554/0.29 7/0.819; rs2895795 TT/AT/AA) loci of three genotypes in three there was no significant difference between groups (P=0.105/0.104/0.730); rs1042718 genotype had no statistically significant difference between the three groups (P=0.141/0.051/0.846). Conclusion: ADRB2 gene polymorphism and uncontrolled asthma had no significant correlation, which may be related to racial differences in haplotype.
