‘云香’水仙NtWRKYY1基因的分離與功能分析

【摘要】:為明確WRKY轉(zhuǎn)錄因子在‘云香’水仙中的功能,該研究以‘云香’水仙為材料,克隆了WRKY基因,命名為NtWRKYY1(GenBank登錄號(hào)為KX056495)。序列分析顯示,NtWRKYY1基因開放閱讀框(ORF)長(zhǎng)度為510bp,編碼169個(gè)氨基酸。多序列比對(duì)和系統(tǒng)進(jìn)化樹分析顯示,NtWRKYY1編碼蛋白含1個(gè)WRKY結(jié)構(gòu)域和C2H2鋅指結(jié)構(gòu)(Cx4Cx23HxH),與AtWRKY57聚在一起,屬于第Ⅱc類WRKY轉(zhuǎn)錄因子。組織表達(dá)和時(shí)空表達(dá)分析顯示,NtWRKYY1基因在花中表達(dá)量高于根和葉,且在花瓣及副冠中的表達(dá)量隨開花過(guò)程(花蕾期、始花期、盛花期、衰敗期)呈上升趨勢(shì)。植物激素和非生物脅迫分析顯示,NtWRKYY1基因受脫落酸(ABA)、高溫、干旱和鹽誘導(dǎo),受茉莉酸甲酯(JA)抑制,表明NtWRKYY1基因可能在‘云香’水仙花朵的衰老過(guò)程中起正調(diào)節(jié)作用,同時(shí)參與‘云香’水仙ABA、JA等激素信號(hào)轉(zhuǎn)導(dǎo)及高溫、干旱、鹽堿等非生物脅迫過(guò)程的調(diào)控。利用In-Fusion克隆技術(shù)成功構(gòu)建過(guò)表達(dá)載體pMDC140-NtWRKYY1,并采用農(nóng)桿菌介導(dǎo)葉盤法轉(zhuǎn)化煙草。RT-PCR和GUS染色結(jié)果顯示,目的基因已成功導(dǎo)入煙草基因組中。
[Abstract]:In order to clarify the function of WRKY transcription factors in Narcissus sinensis, the WRKY gene was cloned from Narcissus sinensis and named NtWRKYY1(GenBank accession number KX056495.Sequence analysis showed that the ORF length of NtWRKYY1 gene was 510bp, encoding 169 amino acids.Multiple sequence alignment and phylogenetic tree analysis showed that the NtWRKYY1 encoded protein contained a WRKY domain and a C2H2 zinc finger structure, Cx4Cx23HxHX, which were clustered with AtWRKY57 and belonged to WRKY transcription factor of class 鈪,
