金屬脅迫下靈芝寡肽轉(zhuǎn)運(yùn)蛋白基因家族的轉(zhuǎn)錄表達(dá)

【摘要】:【目的】寡肽轉(zhuǎn)運(yùn)蛋白(Oligopeptide transporters,OPTs)通過細(xì)胞質(zhì)膜,將細(xì)胞外環(huán)境中的物質(zhì)轉(zhuǎn)入細(xì)胞內(nèi),從而參與各種生理活動。植物中OPTs蛋白參與金屬運(yùn)輸和分配,從而促進(jìn)植物對金屬的利用和適應(yīng),但真菌中OPTs是否參與這一過程的相關(guān)報道較少。探究OPTs是否在靈芝對金屬的適應(yīng)過程中發(fā)揮作用。【方法】以靈芝榮保1號為材料,利用平板實驗分析不同濃度的Cu2+、Fe2+和Se4+對菌絲體生長和生物量的影響,并采用實時熒光定量PCR分析不同金屬離子脅迫下,兩個培養(yǎng)階段(15 d和30 d)靈芝寡肽轉(zhuǎn)運(yùn)蛋白基因(OPTs)的轉(zhuǎn)錄表達(dá)水平!窘Y(jié)果】Cu2+、Fe2+和Se4+三個金屬離子在實驗使用的濃度范圍內(nèi)抑制靈芝菌絲體的生長量和生物量,且在相同處理條件下的生長量與生物量的變化趨勢相同。熒光定量PCR結(jié)果顯示,除未檢測到轉(zhuǎn)錄本的3個基因(OPT7、OPT8和OPT9),其余10個基因均表現(xiàn)出了差異性表達(dá)模式。在培養(yǎng)前期(15 d),有6個寡肽轉(zhuǎn)運(yùn)蛋白基因(OPT1、OPT3-6和OPT10)的相對表達(dá)量在Cu2+的脅迫下發(fā)生上調(diào),而在Fe2+和Se4+的脅迫下,所有的OPTs基因的表達(dá)量均被下調(diào)。在培養(yǎng)后期(30 d),Se4+脅迫下的OPTs基因的表達(dá)量仍舊被下調(diào),而Cu2+和Fe2+的脅迫下表達(dá)量均顯著上調(diào)!窘Y(jié)論】金屬能夠在不同的程度上影響靈芝的生長量和生物量,同時寡肽轉(zhuǎn)運(yùn)蛋白基因?qū)@些金屬脅迫在轉(zhuǎn)錄水平上做出了響應(yīng),表明寡肽轉(zhuǎn)運(yùn)蛋白基因可能在靈芝對金屬的適應(yīng)過程中發(fā)揮作用。
[Abstract]:[objective] Oligopeptide transporters OPTs (Oligopeptide transportersOPTs) were transfered into the cells through the cytoplasmic membrane and involved in various physiological activities.OPTs proteins in plants are involved in metal transport and distribution, thus promoting the utilization and adaptation of plants to metals. However, there are few reports on whether OPTs is involved in this process in fungi.To explore whether OPTs plays a role in the adaptation of Ganoderma lucidum to metal. [methods] the effects of Cu2 Fe2 and Se4 on mycelium growth and biomass were studied by using Ganoderma lucidum Rong Bao 1 as a material.The real-time fluorescence quantitative PCR was used to analyze the stress of metal ions.Transcriptional expression level of Ganoderma lucidum oligoseptide transporter gene (OPTs) was observed at 15 d and 30 d after incubation. [results] Cu2 Fe2 and Se4 inhibited the growth and biomass of Ganoderma lucidum mycelium in the concentration range used in the experiment.And under the same treatment conditions, the change trend of biomass and growth is the same.Fluorescence quantitative PCR showed that all the other 10 genes showed different expression patterns except for 3 genes (OPT7OPT8 and OPT9G) which were not detected in transcripts.The relative expression of OPT1OPT3-6 and OPT10) was up-regulated under the stress of Cu2, but the expression of all OPTs genes was down-regulated under the stress of Fe2 and Se4.The expression of OPTs gene was still down-regulated at 30 days after culture, while that of Cu2 and Fe2 increased significantly. [conclusion] Metal can affect the growth and biomass of Ganoderma lucidum in different degree.At the same time, the oligoseptide transporter genes are responsive to these metal stresses at the transcriptional level, suggesting that the oligopeptide transporter genes may play a role in the adaptation of Ganoderma lucidum to metals.
