南瓜肌醇單磷酸酶基因CmIMP1和CmIMP2的克隆與表達(dá)分析

【摘要】:以中國(guó)南瓜(Cucurbita moschata)為試驗(yàn)材料,利用同源克隆和南瓜轉(zhuǎn)錄組unigene序列獲得2個(gè)IMP基因的全長(zhǎng)c DNA序列,將兩個(gè)基因命名為Cm IMP1和Cm IMP2,Gen Bank登錄號(hào)為KP735607和KP735608。Cm IMP1基因c DNA全長(zhǎng)1 053 bp,包含1個(gè)810 bp的ORF,共編碼269個(gè)氨基酸;Cm IMP2基因c DNA全長(zhǎng)945 bp,包含1個(gè)807 bp的ORF,共編碼268個(gè)氨基酸。序列分析發(fā)現(xiàn)兩個(gè)基因均含有磷酸酶家族鋰敏感的3個(gè)特殊結(jié)構(gòu)域,系統(tǒng)進(jìn)化樹分析表明這2個(gè)南瓜Cm IMP與其他植物IMP有較高同源性(63.8%~94.0%),并與葫蘆科作物最接近。采用熒光定量PCR研究Cm IMP在各組織及逆境條件下的表達(dá)模式顯示,其表達(dá)具有組織特異性,在葉中表達(dá)最高,須和幼果次之;鹽脅迫和干旱脅迫可強(qiáng)烈誘導(dǎo)Cm IMP表達(dá),ABA對(duì)Cm IMP的誘導(dǎo)較微弱,推測(cè)Cm IMP在南瓜響應(yīng)非生物脅迫的分子調(diào)控機(jī)制方面發(fā)揮重要作用。
[Abstract]:Using Cucurbita moschata (Cucurbita moschata) as experimental material, two full-length c DNA sequences of IMP gene were obtained by homologous cloning and unigene sequence of pumpkin transcriptome.Sequence analysis showed that the two genes contained three special domains sensitive to lithium phosphatase family. Phylogenetic tree analysis showed that the two pumpkin cm IMP had a high homology with other plant IMP, 63.80.It was the closest to Cucurbitaceae crops.Fluorescence quantitative PCR was used to study the expression patterns of cm IMP in different tissues and under different stress conditions. The results showed that the expression of cm IMP was tissue specific, the highest in leaves, and the second in young fruits.Salt stress and drought stress could strongly induce the expression of cm IMP on cm IMP, suggesting that cm IMP plays an important role in the molecular regulation of pumpkin response to abiotic stress.
