白三葉新品系抗寒生理特性及其相關(guān)基因表達(dá)研究

【摘要】:本文以白三葉(Trifolium repens.L.)新品系、海法白三葉(Trifolium repe L.cv.haifa),雷司令白三葉(Trifolium repens L.cv.Riesling)為試驗(yàn)材料,研究各白三葉材料種子萌發(fā)和萌發(fā)期、幼苗期、越冬期其生理指標(biāo)的變化,和幼苗期基因MYB4、CBF3、CAS5在葉片中的相對(duì)表達(dá)量的多少,從而了解各白三葉材料的抗寒性能力的強(qiáng)弱,為深入研究白三葉的抗寒機(jī)制和培育抗寒新品種奠定了理論基礎(chǔ)。本研究通過(guò)方差分析、相關(guān)性分析、隸屬函數(shù)綜合分析法處理數(shù)據(jù),得出以下結(jié)論:1.白三葉新品系種子萌發(fā)期和幼苗期抗寒性顯著高于海法白三葉和雷司令白三葉2個(gè)對(duì)照品種。低溫脅迫下,白三葉新品系種子發(fā)芽率和發(fā)芽指數(shù)均高于對(duì)照品種,種子萌發(fā)期在溫度脅迫下超氧化物歧化酶SOD活性、可溶性糖SS、可溶性蛋白SP含量顯著高于對(duì)照品種。生長(zhǎng)期幼苗在5℃低溫脅迫下也有相同的生理變化。2.在自然越冬過(guò)程中,白三葉新品系的抗寒性顯著高于對(duì)照品種。隨著溫度下降,白三葉新品系根部POD、SOD、CAT、SS、SP、PRO的含量均顯著高于對(duì)照品種,其中PRO含量在低溫脅迫下增加量最大;而MDA、REC的含量均顯著低于對(duì)照品種。3.MYB4、CBF3和CAS5基因均受低溫脅迫誘導(dǎo)表達(dá),且在低溫脅迫過(guò)程中呈現(xiàn)一定的變化規(guī)律。其中CBF3基因隨低溫脅迫時(shí)間增加,表達(dá)量持續(xù)上升,MYB4、CAS5在溫脅迫過(guò)程中表達(dá)量先增加后減少;這3個(gè)基因在白三葉新品系中的表達(dá)量均高于海法白三葉和雷司令白三葉,且CBF3基因的表達(dá)量最高,MYB4最低。研究結(jié)果從抗寒相關(guān)功能基因的表達(dá)特性方面初步揭示了白三葉新品系抗逆御寒分子機(jī)制。
[Abstract]:In this paper, a new strain of Trifolium repens.L.), Trifolium repe L.cv. Haifaa and Trifolium repens L.cv. Rieslingn) were used as experimental materials to study the changes of physiological indexes in seed germination and germination period, seedling stage and overwintering stage of white clover. And the relative expression of MYB4 CBF3CAS5 gene in leaves, so as to understand the cold resistance of white clover materials. This study laid a theoretical foundation for the further study of the cold resistance mechanism of white clover and the cultivation of new cold resistant varieties. The data were processed by means of variance analysis, correlation analysis and membership function comprehensive analysis. The following conclusions were drawn: 1. The cold resistance of the new white clover line was significantly higher than that of Haifa white clover and Riesling white clover at germination and seedling stage. The seed germination rate and germination index of the new white clover line were higher than that of the control variety, and the SOD activity of superoxide dismutase (SOD) in the seed germination period was higher than that of the control. The content of soluble sugar SSand soluble protein SP was significantly higher than that of the control variety, and there were the same physiological changes under 5 鈩,
