茶樹β-淀粉酶基因CsBAM3的克隆及其響應(yīng)低溫的表達(dá)模式

【摘要】:β-淀粉酶(BAM)是植物中參與淀粉水解的關(guān)鍵酶類,在應(yīng)對非生物脅迫中發(fā)揮重要作用。從前期茶樹冷馴化轉(zhuǎn)錄組分析中分離到1個參與淀粉代謝的差異表達(dá)基因,cDNA全長克隆及序列分析鑒定該基因?yàn)閿M南芥BAM3同源基因,命名為CsBAM3。該基因編碼548個氨基酸殘基,與擬南芥的BAM1和BAM3一起歸為第Ⅱ亞家族,推測為葉綠體定位、具有淀粉水解活性的β-淀粉酶編碼基因。啟動子克隆及序列分析顯示該基因可能受生理節(jié)律、光、低溫及多種激素等信號共同調(diào)控。CsBAM3在葉片中表達(dá)量最高,莖和花中表達(dá)量較低,根中基本不表達(dá)。CsBAM3在冷馴化初期被顯著上調(diào)且一直保持相對較高的水平。成熟葉片及嫩芽(一芽二葉)中的CsBAM3均可以被4℃及0℃低溫顯著上調(diào),且嫩芽中基因的表達(dá)量上調(diào)幅度明顯高于成熟葉。模擬倒春寒低溫條件下,檢測不同萌發(fā)階段新梢中CsBAM3的表達(dá)情況表明,CsBAM3在鮮葉初展的嫩芽中即可快速受低溫誘導(dǎo)。以上結(jié)果表明,CsBAM3是茶樹中調(diào)控淀粉水解的一個重要β淀粉酶編碼基因,在茶樹成熟葉和嫩芽受到不同低溫脅迫時其表達(dá)可被快速誘導(dǎo)。
[Abstract]:尾 -amylase (尾 -amylase) is the key enzyme involved in starch hydrolysis in plants. A differentially expressed gene involved in starch metabolism was cloned and sequenced to identify the homologous gene of Arabidopsis thaliana BAM3 in Arabidopsis thaliana. CsBAM3.The gene encodes 548 amino acid residues and belongs to the second subfamily along with BAM1 and BAM3 of Arabidopsis thaliana. 尾 -amylase encoding gene with starch hydrolysis activity. Cloning and sequence analysis of promoter showed that the gene may be regulated by physiological rhythm, light, hypothermia and multiple hormones. CsBAM3 is the most expressed gene in leaves. The expression of CsBAM3 in stem and flower was low, the expression of .CsBAM3 in root was significantly up-regulated at the beginning of cold acclimation and maintained a relatively high level.
