鳳丹油體膜蛋白基因PoOLE17.5的克隆與表達特性分析

【摘要】:油體是油料作物種子儲存脂肪的細胞器,油體膜蛋白是構(gòu)成油體的重要組分,對維持油體大小和穩(wěn)定起著重要作用。本研究以油體膜蛋白為對象,克隆了鳳丹的油體膜蛋白基因(OLE),其cDNA全長783 bp,開放閱讀框長507 bp,共編碼169個氨基酸,蛋白的分子量為17.48 k D,故將該基因命名為PoOLE17.5。生物信息學分析顯示,該基因編碼的蛋白包含2個親水性區(qū)域和1個疏水性區(qū)域,并具有1個保守結(jié)構(gòu)域"Oleosin domain"。而后的表達模式顯示,PoOLE17.5在鳳丹種子發(fā)育過程中的表達水平逐漸上升,在種子接近成熟時表達水平最高,而后略有下降。這一結(jié)果有利于今后通過基因工程手段調(diào)控鳳丹油體發(fā)育。
[Abstract]:Oil body is the organelle of oil plant seed to store fat. Oil body membrane protein is an important component of oil body and plays an important role in maintaining oil body size and stability. The oil body membrane protein (Ole) gene of Fengdan was cloned. Its cDNA length was 783bpand the length of open reading frame was 507bp. it encoded 169 amino acids, and the molecular weight of the protein was 17.48kD. therefore, the gene was named Poole 17.5. Bioinformatics analysis showed that, The protein encoded by the gene contains two hydrophilic and one hydrophobic regions, and has a conserved domain "Oleosin domain". The expression pattern shows that the expression level of PoOLE17.5 increases gradually during the seed development of Fengdan. The expression level was the highest in the seed near maturation, and then decreased slightly. This result is helpful to regulate the oil body development of Fengdan by genetic engineering in the future.
