玉米磷轉(zhuǎn)運蛋白基因ZmPht3;1的克隆和功能分析

【摘要】:Pht3(phosphate transporter 3)磷轉(zhuǎn)運子家族屬于一類低親和力磷轉(zhuǎn)運蛋白,在調(diào)節(jié)植株體內(nèi)磷素的動態(tài)平衡中發(fā)揮重要作用。為了初步探討玉米中Zm Pht3;1基因的結(jié)構(gòu)特征及其磷饑餓的響應(yīng)機制,利用同源克隆的方法從耐低磷玉米自交系Mo17中分離得到Zm Pht3;1基因,并運用實時熒光定量PCR和亞細胞定位的方法對其進行深入研究。結(jié)果表明,Zm Pht3;1的編碼區(qū)全長1 101 bp,編碼366個氨基酸,含有典型的線粒體轉(zhuǎn)運家族(mitochondrial carrier family,MCF)結(jié)構(gòu)特征與6個疏水跨膜結(jié)構(gòu)。熒光定量PCR分析表明,該基因在兩個極端材料的根系與葉片中均有表達,而表達模式差異顯著,在耐低磷玉米自交系Mo17的根系和葉片中表現(xiàn)為缺磷脅迫前期的一般性反應(yīng)和后期的特異性反應(yīng)。轉(zhuǎn)化煙草的亞細胞定位結(jié)果顯示,Zm Pht3;1主要分布于細胞膜上,可能是一個雙親和轉(zhuǎn)運體,在玉米響應(yīng)磷饑餓脅迫過程中發(fā)揮重要的適應(yīng)性調(diào)節(jié)作用。
[Abstract]:Pht3(phosphate transporter 3) P transporter family belongs to a low affinity phosphorous transporter family, which plays an important role in regulating the dynamic balance of phosphorus in plants. The Zm Pht3H1 gene was isolated from low phosphorus tolerant maize inbred line Mo17 by homologous cloning. The method of real-time fluorescence quantitative PCR and subcellular localization was used to study it. The results showed that the coding region of Zm Pht3H1 was 1 101 BP, encoding 366 amino acids. There are typical mitochondrial carrier family MCFs and six hydrophobic transmembrane structures. Fluorescence quantitative PCR analysis showed that the gene was expressed in both the roots and leaves of the two extreme materials, and the expression patterns were significantly different. In the roots and leaves of low phosphorus tolerant maize inbred line Mo17, the general and late specific responses were observed in the early stage of phosphorus deficiency stress and the specific response in the later stage. The subcellular localization results showed that Zm Pht3O1 was mainly distributed on the cell membrane. It may be a parent and transporter, which plays an important role in the adaptive regulation of maize response to phosphorus starvation stress.
