華癸根瘤菌7653R hfq基因突變株的構建及其生物學特性

【摘要】:【目的】研究hfq基因在Mesorhizobium huakuii 7653R抵抗外界不利環(huán)境和共生固氮中的功能特性。【方法】利用pK19mob同源重組方法構建7653R hfq基因的插入失活突變株7653RΔhfq,并構建互補菌株7653RΔhfq-C,對hfq在壓力脅迫和共生固氮中的功能特性進行研究!窘Y果】與野生型7653R相比,突變株7653RΔhfq的生長速率降低,熱激處理后致死率升高;hfq突變影響了7653R中部分sRNA的表達;在4.5%乙醇和50 mmol H_2O_2生長脅迫下,突變株適應性明顯較野生型差。另外,接種突變株的紫云英結瘤能力和固氮酶活性都明顯降低。【結論】hfq基因作為重要的轉錄后調控因子,在7653R抵御外界脅迫環(huán)境和與宿主紫云英的共生固氮中發(fā)揮了重要作用。
[Abstract]:[objective] to study the functional characteristics of hfq gene in Mesorhizobium huakuii 7653R resistance to external adverse environment and symbiotic nitrogen fixation. [methods] the insertion inactivated mutant 7653R 螖 hfq-Cof 7653R hfq gene was constructed by pK19mob homologous recombination method, and the complementary strain 7653R 螖 hfq-Cwas constructed. The functional characteristics in stress stress and symbiotic nitrogen fixation were studied. [results] compared with wild type 7653R, The growth rate of the mutant 7653R 螖 hfq decreased, the mortality increased after heat shock treatment, and the expression of some sRNA in 7653R was affected by the mutation. Under 4.5% ethanol and 50 mmol H_2O_2 growth stress, the adaptability of the mutant was significantly worse than that of the wild type. [conclusion] as an important posttranscriptional regulator, hfq gene plays an important role in the resistance to external stress and the symbiotic nitrogen fixation with host Ziyun. [conclusion] as an important posttranscriptional regulator, hfq gene plays an important role in resistance to external stress and symbiotic nitrogen fixation.
