【摘要】：探討下調(diào)肝腸鈣黏連蛋白(CDH17)基因?qū)σ种颇强啥∧退幮匀宋赴㎝GC-803細(xì)胞增殖的分子機(jī)制,為臨床治療胃癌提供新的治療手段。本研究以人胃癌MGC-803細(xì)胞為研究材料,通過(guò)體外MTT實(shí)驗(yàn)檢測(cè)細(xì)胞活力、流式細(xì)胞術(shù)觀察細(xì)胞增殖與凋亡情況,蛋白免疫印跡檢測(cè)凋亡通路相關(guān)蛋白的表達(dá)水平變化。那可丁藥物處理實(shí)驗(yàn)和MTT實(shí)驗(yàn)表明,下調(diào)CDH17可以提高人胃癌MGC-803細(xì)胞對(duì)那可丁的敏感性;流式細(xì)胞術(shù)觀察結(jié)果表明,下調(diào)CDH17基因促進(jìn)引起細(xì)胞凋亡;蛋白免疫印跡實(shí)驗(yàn)表明線粒體途徑參與那可丁誘導(dǎo)人胃癌MGC-803細(xì)胞凋亡的過(guò)程。綜上所述,下調(diào)CDH17基因通過(guò)提高人胃癌MGC-803細(xì)胞對(duì)那可丁的敏感性,引起線粒體途徑相關(guān)蛋白功能障礙促進(jìn)細(xì)胞凋亡,從而抑制胃癌細(xì)胞的增殖。
[Abstract]:To explore the molecular mechanism of down-regulation of liver-intestinal calmodulin (CDH17) gene on inhibiting the proliferation of nakodin-resistant human gastric cancer MGC-803 cells, and to provide a new therapeutic method for clinical treatment of gastric cancer. In this study, human gastric cancer MGC-803 cells were used as the research materials. In vitro MTT assay was used to detect cell viability, flow cytometry was used to observe cell proliferation and apoptosis, and Western blot was used to detect the expression of apoptosis-related proteins. Down-regulation of CDH17 could increase the sensitivity of MGC-803 cells to nakodin, flow cytometry showed that down-regulation of CDH17 gene promoted apoptosis. Western blot analysis showed that mitochondrial pathway was involved in the apoptosis of human gastric cancer MGC-803 cells induced by nakodin. In summary, down-regulated CDH17 gene enhanced the sensitivity of human gastric cancer MGC-803 cells to nakodin. Mitochondrial pathway related protein dysfunction promotes cell apoptosis and inhibits the proliferation of gastric cancer cells.
