兩種水稻短穗突變體的形態(tài)特征、遺傳分析和基因定位

【摘要】:通過輻射誘變秈稻品種明恢86,獲得兩種短穗突變體,分別命名為rpl1(reduced panicle length 1)和rpl2(reduced panicle length 2)。兩突變體都表現(xiàn)為穗變短、枝梗數(shù)和穗粒數(shù)減少、枝梗縮短,但其再生植株的穗長(zhǎng)有所恢復(fù),與野生型的差異明顯減少。等位性測(cè)驗(yàn)表明,rpl1和rpl2是等位突變。利用M4分離群體和基于高通量測(cè)序的分離體混合分析(BSA-seq)方法,發(fā)現(xiàn)rpl2中LOC_Os11g12740基因的剪接位點(diǎn)發(fā)生了單堿基置換突變。進(jìn)一步對(duì)rpl1測(cè)序分析,發(fā)現(xiàn)該基因完全缺失。這說明rpl1和rpl2都是LOC_Os11g12740突變引起的,由此證實(shí)它就是目標(biāo)基因。該基因與已報(bào)道的SP1是同一個(gè)基因,因此rpl1和rpl2是SP1的新等位突變,但二者的表型與已報(bào)道的3種短穗突變體并不完全相同,說明不同突變和遺傳背景會(huì)影響基因功能的表現(xiàn)。本研究進(jìn)一步驗(yàn)證了SP1的功能,并為深入研究其分子作用機(jī)理提供新材料。
[Abstract]:Two short spike mutants, named rpl1(reduced panicle length 1) and rpl2(reduced panicle length 2, were obtained by radiation-induced transformation of indica rice variety Minghui 86. The two mutants were characterized by shorter panicles, fewer branches and grains and shorter branches. However, the panicle length of regenerated plants recovered, and the difference with wild type was significantly reduced. Allelic test showed that rpl1 and rpl2 were allelic mutations. BSA-seq method was used to isolate M4 population and isolate BSA-seq based on high-throughput sequencing. A single base substitution mutation was found in the splicing site of the LOC_Os11g12740 gene in rpl2. Further analysis of rpl1 sequencing showed that the gene was completely missing, which indicated that both rpl1 and rpl2 were caused by LOC_Os11g12740 mutation. Therefore, rpl1 and rpl2 are new alleles of SP1, but their phenotypes are not identical with the three reported short spike mutants. This study further verifies the function of SP1 and provides a new material for further study of its molecular mechanism.
