【摘要】:為簡化和提高轉(zhuǎn)化效率,以蓖麻品種汾蓖10號(hào)為材料,采用非組培的農(nóng)桿菌介導(dǎo)萌動(dòng)種胚轉(zhuǎn)化法,獲得抗草甘膦植株T0。方法是在生長點(diǎn)附近針刺3針,超聲波(強(qiáng)度800W)處理后,與農(nóng)桿菌共培養(yǎng)16h。在隨機(jī)選取的800株轉(zhuǎn)化處理植株中,檢測到5株P(guān)CR陽性植株,平均轉(zhuǎn)化率為6.3‰;在噴施除草劑后存活的26株轉(zhuǎn)化處理植株中,經(jīng)PCR和Southern blot檢測,獲得了3株陽性抗草甘膦T0植株。本方法操作簡單,成本低,避免了冗長繁瑣的組織培養(yǎng)過程,為蓖麻遺傳轉(zhuǎn)化研究提供了新的思路和方法。
[Abstract]:In order to simplify and improve the transformation efficiency, the ricin variety Fen-gri 10 was used to obtain glyphosate resistant plants T0 by non-tissue culture Agrobacterium tumefaciens mediated embryo transformation. After 16 h co-culture with Agrobacterium tumefaciens, 5 PCR positive plants were detected in 800 randomly selected transformed plants with an average conversion rate of 6.3 鈥,
