【摘要】：為了研究PIWI基因?qū)d羊生殖細(xì)胞發(fā)育的影響,試驗采用同源基因克隆的方法,以綿羊睪丸、卵巢RNA為模板,通過PCR擴(kuò)增,半定量PIWI基因家族在蘇尼特羊、小尾寒羊生殖腺的表達(dá)。結(jié)果表明:公羊PIWI蛋白家族3個成員mRNA的相對表達(dá)水平均極顯著高于母羊(P0.01),PIWI1、PIWI2、PIWI4在高繁小尾寒羊母羊、低繁蘇尼特母羊均只有微弱表達(dá),同性別高繁小尾寒羊、低繁蘇尼特羊mRNA相對表達(dá)水平差異不顯著(P0.05)。
[Abstract]:In order to study the effect of PIWI gene on the development of ovine germ cells, the homologous gene cloning method was used to amplify RNA from testis and ovaries of sheep. The semi-quantitative PIWI gene family was amplified by PCR in Sunita sheep. The results showed that the relative expression level of mRNA in the three members of the PIWI protein family of male sheep was significantly higher than that of female sheep P0.01PIWI2PIWI2PIWI4, and only weak expression of PIWI2PIWI4 was found in the high and small tail Han sheep, and in the low breed Sunita ewe, the expression level of mRNA was significantly higher than that of P0.01PIWI2 and PIWI2PIWI4. There was no significant difference in the relative expression level of mRNA between small tail Han sheep and low procreation Sunita sheep.
