【摘要】：Aux/IAA(auxin/indole-3-acetic acid)作為生長素早期響應因子,其蛋白產(chǎn)物能夠特異性地結合生長素響應因子(auxin response factor,ARF),從而調控生長素響應基因的表達,在整個植物生長素信號轉導過程中發(fā)揮著重要的作用,進而影響植物的生長發(fā)育。本研究通過構建沉默載體,在高效遺傳轉化體系下獲得轉基因植株,發(fā)現(xiàn)利用Gateway克隆技術將番茄生長素早期響應基因Sl IAA16 DomainⅡ區(qū)部分序列連入具有正反2個attr區(qū)域并連接1個內含子的p B7GWIWG2(Ⅰ)載體,可在植株體內形成發(fā)卡結構導致植物基因沉默,成功構建基因沉默表達載體名為Sl IAA RNAi;此外,抗生素噴施和PCR擴增以及實時定量檢測進一步鑒定,共獲得12棵Sl IAA16 RNAi陽性植株。本研究結果將為進一步明確Sl IAA的生物學功能和闡明生長素的作用機理提供參考。
