番茄SlIAA16沉默轉(zhuǎn)基因植株的獲得及功能初探

【摘要】:Aux/IAA(auxin/indole-3-acetic acid)作為生長(zhǎng)素早期響應(yīng)因子,其蛋白產(chǎn)物能夠特異性地結(jié)合生長(zhǎng)素響應(yīng)因子(auxin response factor,ARF),從而調(diào)控生長(zhǎng)素響應(yīng)基因的表達(dá),在整個(gè)植物生長(zhǎng)素信號(hào)轉(zhuǎn)導(dǎo)過(guò)程中發(fā)揮著重要的作用,進(jìn)而影響植物的生長(zhǎng)發(fā)育。本研究通過(guò)構(gòu)建沉默載體,在高效遺傳轉(zhuǎn)化體系下獲得轉(zhuǎn)基因植株,發(fā)現(xiàn)利用Gateway克隆技術(shù)將番茄生長(zhǎng)素早期響應(yīng)基因Sl IAA16 DomainⅡ區(qū)部分序列連入具有正反2個(gè)attr區(qū)域并連接1個(gè)內(nèi)含子的p B7GWIWG2(Ⅰ)載體,可在植株體內(nèi)形成發(fā)卡結(jié)構(gòu)導(dǎo)致植物基因沉默,成功構(gòu)建基因沉默表達(dá)載體名為Sl IAA RNAi;此外,抗生素噴施和PCR擴(kuò)增以及實(shí)時(shí)定量檢測(cè)進(jìn)一步鑒定,共獲得12棵Sl IAA16 RNAi陽(yáng)性植株。本研究結(jié)果將為進(jìn)一步明確Sl IAA的生物學(xué)功能和闡明生長(zhǎng)素的作用機(jī)理提供參考。
