枸杞花藥發(fā)育基因LbMS2的克隆及表達(dá)分析

【摘要】:MS2(Male sterily2)類基因編碼脂肪;原酶,參與了花藥絨氈層中脂類代謝過程。該研究以寧夏枸杞(‘寧杞1號’)為試驗材料,采用RACE方法從枸杞花藥cDNA中獲得枸杞LbMS2基因。結(jié)果顯示,LbMS2基因開放閱讀框為1 782bp,編碼593個氨基酸,等電點為8.98;生物信息學(xué)分析顯示,LbMS2蛋白定位于葉綠體;LbMS2蛋白序列與茄科植物矮牽牛、茸毛煙草、馬鈴薯、番茄以及甜辣椒中的MS2蛋白表現(xiàn)出較高的序列相似性;實時熒光定量PCR結(jié)果顯示,LbMS2基因具有器官表達(dá)特異性,只在枸杞花器官中表達(dá),并且在枸杞花藥發(fā)育的四分體時期以及單核花粉時期表達(dá)量最高。研究表明,LbMS2基因是枸杞花器官發(fā)育過程中的重要基因。
[Abstract]:MS2(Male sterily2) gene encodes fatty acyl reductase, which is involved in lipid metabolism in anther tapetum. The LbMS2 gene of Lycium barbarum L. was obtained from Chinese wolfberry anther cDNA by RACE method. The results showed that the open reading frame of LbMS2 gene was 1782 BP, encoding 593 amino acids. The isoelectric point was 8.98. Bioinformatics analysis showed that the sequence of LbMS2 protein located in chloroplast was similar to that in petunia, furry tobacco, potato, tomato and sweet capsicum, and the sequence of LbMS2 protein was similar to that in eggplant petunia, furry tobacco, potato, tomato and sweet pepper. The results of real-time fluorescence quantitative PCR showed that LbMS2 gene had organ expression specificity and was only expressed in flower organs of Lycium barbarum L. The results showed that LbMS2 gene was an important gene in the flower organ development of Lycium barbarum L.
