【摘要】：為確定引起廣東環(huán)尾狐猴(Lemur catta)疑似弓形蟲(chóng)病的病原,從2只死亡的環(huán)尾狐猴體內(nèi)無(wú)菌采集肺臟、肝臟、心、腎和腦等組織制作常規(guī)石蠟切片,HE染色。通過(guò)病理組織學(xué)觀察,鑒定為弓形蟲(chóng)(Toxoplasma sp.)后,采用PCR-DNA測(cè)序分析方法對(duì)環(huán)尾狐猴源分離蟲(chóng)株(TgRTL1)的SAG1、SAG2、SAG3、BTUB、GRA6、L358、PK1、c22-8、c29-2和Apico基因位點(diǎn)進(jìn)行PCR擴(kuò)增、克隆和測(cè)序,采用DNAStar和MEGA5.05軟件,將所獲序列與網(wǎng)上下載的弓形蟲(chóng)RH株、ME49株和VEG株相應(yīng)序列進(jìn)行比對(duì)和同源性分析,并構(gòu)建系統(tǒng)發(fā)育樹(shù)。結(jié)果顯示,從廣東分離的環(huán)尾狐猴源弓形蟲(chóng)株,在10個(gè)遺傳位點(diǎn)經(jīng)PCR-DNA測(cè)序分析,確定該蟲(chóng)株的基因型為Toxo DB#3。本研究是對(duì)中國(guó)環(huán)尾狐猴源弓形蟲(chóng)采用多位點(diǎn)DNA序列分析進(jìn)行基因型鑒定的首次報(bào)道,為進(jìn)一步研究我國(guó)弓形蟲(chóng)流行病學(xué)、生物學(xué)特性以及弓形蟲(chóng)病的診斷提供了科學(xué)理論依據(jù)。
[Abstract]:In order to determine the pathogen of the suspected toxoplasmosis, lung, liver, heart, kidney and brain tissues were collected from two dead lemurs to make routine paraffin sections and HE staining. After being identified as Toxoplasma sp.), SAG1, SAG2 and SAG3 of TgRTL1) were amplified by PCR, cloned and sequenced by PCR, cloned and sequenced by DNAStar and MEGA5.05 software. The sequence was compared with the corresponding sequences of Toxoplasma gondii RH strain ME49 and VEG strain, and the phylogenetic tree was constructed. The genotypes of Toxoplasma gondii were determined to be Toxo DB#3 by PCR-DNA sequencing analysis at 10 genetic loci. This study was the first report to identify the genotypes of Toxoplasma gondii (Toxoplasma gondii) by multilocus DNA sequence analysis. It provides scientific theoretical basis for further study on epidemiology, biological characteristics and diagnosis of Toxoplasma gondii in China.
