【摘要】：目的觀察加味牛蒡子湯(JWNBZT)對(duì)類風(fēng)濕關(guān)節(jié)炎(RA)模型動(dòng)物TNF-α轉(zhuǎn)基因(TNF-Tg)小鼠關(guān)節(jié)炎癥和關(guān)節(jié)周圍淋巴回流功能的影響。方法將16只清潔級(jí)3月齡左右TNF-Tg小鼠按照體重隨機(jī)分為TNF-Tg組和JWNBZT組,每組8只。另取10只同窩對(duì)照體重相似的野生型小鼠(WT)作為對(duì)照組,JWNBZT組每天以JWNBZT灌胃,對(duì)照組和TNF-Tg組每天以等量純凈水灌胃,治療12周后小鼠足底皮下注射吲哚菁綠(ICG)測(cè)量小鼠淋巴管回流功能,并取踝關(guān)節(jié)制作石蠟切片進(jìn)行蘇木素-伊紅(HE)染色觀察炎性浸潤(rùn)和關(guān)節(jié)骨質(zhì)破壞情況,抗酒石酸酸性磷酸酶(TRAP)染色觀察破骨細(xì)胞數(shù)量。結(jié)果與對(duì)照組比較,TNF-Tg組小鼠淋巴管回流功能出現(xiàn)明顯損害,Clearance和Pulse明顯下降,淋巴管滲漏面積增多,踝關(guān)節(jié)內(nèi)出現(xiàn)明顯炎癥浸潤(rùn)且骨質(zhì)減少,距骨內(nèi)破骨細(xì)胞增多(P0.05);與TNF-Tg組比較,JWNBZT組小鼠淋巴管回流功能明顯增強(qiáng),Clearance和Pulse升高,淋巴管滲漏面積減少,踝關(guān)節(jié)內(nèi)炎癥浸潤(rùn)減輕且骨質(zhì)增多,距骨內(nèi)破骨細(xì)胞減少(P0.05)。結(jié)論 JWNBZT能夠減輕RA受累關(guān)節(jié)的炎癥浸潤(rùn)和關(guān)節(jié)骨質(zhì)破壞,并促進(jìn)淋巴管回流功能,這可能是其治療RA的機(jī)制。
[Abstract]:Objective to observe the effect of JWNBZT (JWNBZT) on arthritis and peripheral lymphoid reflux in TNF- 偽 transgenic TNF-Tg mice. Methods Sixteen TNF-Tg mice of 3 months old of clean grade were selected to follow their body weight according to their body weight. The machine was divided into TNF-Tg group and JWNBZT group. Eight mice in each group were taken as control group, and 10 wild type mice with similar body weight were taken as control group. JWNBZT group was treated with JWNBZT daily, and the control group and TNF-Tg group were treated with the same amount of pure water daily. After 12 weeks of treatment, the subcutaneous injection of indocyanine green (ICG) into the plantar of mice was used to measure the function of lymphatic reflux in mice, and paraffin sections were taken from the ankle joint to be stained with hematoxylin and eosin (HEH) to observe the inflammatory infiltration and joint bone destruction. The number of osteoclasts was observed by tartrate-resistant acid phosphatase (TRAP) staining. Results compared with the control group, the lymphatic reflux function of TNF-Tg group decreased significantly, and the lymphatic vessel leakage area increased. Significant inflammatory infiltration and osteopenia were observed in the ankle joint, and osteoclasts increased in the talus (P 0.05). Compared with the TNF-Tg group, the lymphatic reflux function of the JWNBZT group was significantly increased, and the lymphatic vessel leakage area was decreased in the JWNBZT group, and the function of lymphatic reflux was significantly increased in the JWNBZT group. Conclusion JWNBZT can reduce inflammatory infiltration and joint bone destruction and promote lymphatic reflux function, which may be the mechanism of JWNBZT in the treatment of RA.
