在吸水鏈霉菌中表達(dá)透明顫菌血紅蛋白基因提高子囊霉素產(chǎn)量

【摘要】:為了解決子囊霉素(1)發(fā)酵過(guò)程低溶氧的問(wèn)題,本研究將透明顫菌(Vitreoscilla)血紅蛋白基因(vgb)整合至1生產(chǎn)菌株吸水鏈霉菌(Streptomyces hygroscopicus)中,構(gòu)建工程菌SIPI-FK520-2,通過(guò)RT-PCR分析檢測(cè)到了vgb基因的轉(zhuǎn)錄表達(dá),進(jìn)一步利用CO結(jié)合差光譜分析也證實(shí)了工程菌中透明顫菌血紅蛋白(VHb)具有生物活性。最后在發(fā)酵罐進(jìn)行該工程菌的發(fā)酵培養(yǎng)研究,結(jié)果表明,與野生菌相比,工程菌發(fā)酵過(guò)程溶氧水平明顯改善,1產(chǎn)量提高了38%,達(dá)到1 269.7 mg/L。
[Abstract]:In order to solve the problem of low dissolved oxygen during ascomycin 1) fermentation, the hemoglobin gene of Vitreoscilla (VGB) was integrated into Streptomyces hygroscopicus. the engineering strain SIPI-FK520-2 was constructed, and the transcriptional expression of vgb gene was detected by RT-PCR analysis. Furthermore, the biological activity of VHb was confirmed by CO binding differential spectrum analysis. Finally, the fermentation culture of the engineered strain was studied in fermenter. The results showed that compared with wild bacteria, VHb was more bioactive than that of wild bacteria. The dissolved oxygen level in the fermentation process of engineering bacteria obviously improved the yield of T1 by 38%, reaching 12269.7 mg / L 路L ~ (-1) 路L ~ (-1) 路L ~ (-1).
