保定地區(qū)奶牛源隱孢子蟲(chóng)的分離與基因鑒定

【摘要】:為了解保定地區(qū)奶牛源隱孢子蟲(chóng)感染情況,從當(dāng)?shù)?個(gè)奶牛場(chǎng)隨機(jī)采集145份奶牛糞便經(jīng)飽和蔗糖溶液漂浮后直接鏡檢和抗酸染色后鏡檢,陽(yáng)性樣品采用PCR方法擴(kuò)增18S rRNA基因,同時(shí)將擴(kuò)增出的目的片段進(jìn)行克隆和序列分析,并將分離株與其他11個(gè)隱孢子蟲(chóng)參考株的18S rRNA序列進(jìn)行比對(duì)和遺傳距離比較。結(jié)果表明:有10份糞便樣本為隱孢子蟲(chóng)陽(yáng)性,感染率為6.9%(10/145),不同奶牛場(chǎng)、年齡段奶牛隱孢子蟲(chóng)感染率有顯著性差異(P0.05)。10份糞便樣品采用PCR方法擴(kuò)增后有7份為18S rRNA基因陽(yáng)性,擴(kuò)增出的18S rRNA部分基因片段長(zhǎng)528 bp。保定地區(qū)隱孢子蟲(chóng)分離株擴(kuò)增的基因序列與牛源隱孢子蟲(chóng)18S rRNA基因序列(Gen Bank登錄號(hào)為HQ179571)同源性最高,確定保定地區(qū)分離到的奶牛源隱孢子蟲(chóng)為牛隱孢子蟲(chóng)。
[Abstract]:In order to understand the infection of Cryptosporidium from dairy cattle in Baoding area, 145 samples of cow feces were collected randomly from three dairy farms in Baoding. The 18s rRNA gene was amplified by PCR method after floating in saturated sucrose solution and acid fast staining. The amplified fragments were cloned and sequenced. The 18s rRNA sequences of the isolates were compared with those of the other 11 Cryptosporidium reference strains. The results showed that 10 fecal samples were Cryptosporidium positive. The infection rate of Cryptosporidium in different dairy farms was significantly different from that of 10 / 145g in dairy cattle. Seven of the 10 fecal samples amplified by PCR were 18s rRNA positive, and the infection rate of Cryptosporidium was significantly higher than that of the control, and there was significant difference in the infection rate of Cryptosporidium in dairy cattle. The amplified partial gene fragment of 18s rRNA was 528bp.The amplified gene sequence of Cryptosporidium isolated from Baoding area was the highest homology with the accession number HQ179571 of the 18s rRNA gene sequence of Cryptosporidium bovis (HQ179571). Cryptosporidium was identified as Cryptosporidium bovis in Baoding area.
