水牛甲狀旁腺素基因克隆與生物信息學(xué)分析

【摘要】:本研究參考黃牛甲狀旁腺素(parathyroid hormone,PTH)基因序列設(shè)計(jì)引物,運(yùn)用PCR擴(kuò)增技術(shù)對(duì)水牛PTH基因進(jìn)行克隆,并用生物信息學(xué)方法對(duì)序列進(jìn)行分析。結(jié)果顯示,成功克隆了水牛PTH基因完整編碼區(qū)序列(CDS)348bp,可編碼115個(gè)氨基酸。該基因編碼區(qū)序列與黃牛、豬、馬、山羊、綿羊和駱駝的同源性分別為99%、93%、91%、97%、96%和92%,表明PTH基因在不同物種間高度保守。系統(tǒng)進(jìn)化樹(shù)分析表明,水牛與黃牛PTH基因親緣關(guān)系最近。此外,在水牛PTH基因CDS上共發(fā)現(xiàn)了3個(gè)堿基突變,其中兩個(gè)屬于同義突變,一個(gè)屬于錯(cuò)義突變。氨基酸序列分析表明,該蛋白屬于堿性、親水、穩(wěn)定型蛋白質(zhì),其分子式為C_(570)H_(941)N_(167)O_(164)S_8,分子質(zhì)量為13.01ku。二級(jí)結(jié)構(gòu)分析顯示,水牛PTH蛋白以α-螺旋和無(wú)規(guī)則卷曲為主,與三級(jí)結(jié)構(gòu)預(yù)測(cè)結(jié)果相一致。亞細(xì)胞定位分析顯示,水牛PTH蛋白分布在細(xì)胞質(zhì)(26.1%)、線(xiàn)粒體(21.7%)、細(xì)胞外(包括細(xì)胞壁)(17.4%)、內(nèi)質(zhì)網(wǎng)(13.0%)、細(xì)胞核(8.7%)、高爾基體(4.3%)和其他部位(8.7%),推測(cè)PTH蛋白可能在運(yùn)輸、結(jié)合及細(xì)胞被膜等方面發(fā)揮信號(hào)轉(zhuǎn)導(dǎo)和轉(zhuǎn)錄調(diào)控作用。
[Abstract]:In this study, we designed primers for the sequence of parathyroid hormoney (PTH) gene of bovine parathyroid hormone, cloned the PTH gene of buffalo by PCR amplification technique, and analyzed the sequence by bioinformatics. The complete coding region of buffalo PTH gene was cloned successfully, which can encode 115 amino acids. The sequence of the complete coding region of buffalo PTH gene is similar to that of cattle, pig, horse, goat, yellow cattle, pig, horse and goat. The homology of sheep and camel were 9993and 9197%, respectively, indicating that the PTH gene was highly conserved among different species. Phylogenetic tree analysis showed that the PTH gene of buffalo and yellow cattle had the closest relationship. Three base mutations were found in buffalo PTH gene CDS, two of which were synonymous mutations and one was missense mutation. The amino acid sequence analysis showed that the protein belonged to alkaline, hydrophilic and stable proteins. The molecular formula of this protein is: Che 570H / L / N _ S _ (167O) / S _ (8) and molecular mass 13.01ku.The secondary structure analysis shows that the PTH protein of buffalo is mainly 偽 -helix and irregular curl, which is consistent with the prediction result of tertiary structure. The PTH protein of buffalo is distributed in cytoplasm 26.1%, mitochondria 21.7m, extracellular (including cell wall 17.4, endoplasmic reticulum 13.0, nucleus 8.7, Golgi 4.33) and other parts, suggesting that PTH protein may be transported. Binding and cell membrane play a role in signal transduction and transcriptional regulation.
