不同倍性鯽鯉MeCP2基因分子克隆及時(shí)空表達(dá)分析

【摘要】:為了探尋不同倍性魚生殖育性的表觀遺傳調(diào)控規(guī)律,實(shí)驗(yàn)基于不同倍性鯽鯉精巢的轉(zhuǎn)錄本信息庫設(shè)計(jì)引物克隆了不同倍性鯽鯉的MeCP2基因。序列比對結(jié)果發(fā)現(xiàn),在遠(yuǎn)緣雜交的過程中基因組發(fā)生了遺傳重組和變異。基于序列公共部分設(shè)計(jì)引物,通過半定量PCR和實(shí)時(shí)定量PCR的方法分析了MeCP2基因在不同倍性魚發(fā)育過程中的時(shí)空表達(dá)特征。結(jié)果還發(fā)現(xiàn),MeCP2基因在所有組織中都有表達(dá),但在腦組織中的表達(dá)量最高;縱向?qū)Ρ刃韵俚陌l(fā)育過程,MeCP2基因的表達(dá)量會(huì)隨著性腺發(fā)育成熟而下降;橫向?qū)Ρ炔煌缎贼~的性腺發(fā)育,MeCP2基因在三倍體魚卵巢中的表達(dá)量明顯高于二倍體和四倍體魚,但在精巢中的表達(dá)量隨魚的倍性增加而增加。研究表明,MeCP2基因的表達(dá)與不同倍性魚的卵巢發(fā)育密切相關(guān),這為研究魚類生殖不育的分子機(jī)理及其在水產(chǎn)育種上的應(yīng)用奠定了堅(jiān)實(shí)的基礎(chǔ)。
[Abstract]:In order to explore the rules of epigenetic regulation of reproductive fertility of different ploidy fish, primers were designed to clone the MeCP2 gene of different ploidy crucian carp based on the transcripts of testis of different ploidy crucian carp. Genetic recombination and variation occurred in the genome during distant hybridization. Primers were designed based on the common part of the sequence. The temporal and spatial expression characteristics of MeCP2 gene in different ploidy fish were analyzed by semi-quantitative PCR and real-time quantitative PCR. It was also found that MeCP2 gene was expressed in all tissues, but the highest expression level was found in brain tissue. The expression of MeCP2 gene decreased with the development of gonad, and the expression of MeCP2 gene in the ovary of triploid fish was significantly higher than that of diploid and tetraploid fish, and the expression of MeCP2 gene in the gonadal development of different ploidy fish was higher than that of diploid and tetraploid fish. However, the expression level in spermary increased with the increase of fish ploidy. The results showed that the expression of MeCP2 gene was closely related to ovarian development of different ploidy fish. This laid a solid foundation for studying the molecular mechanism of fish reproductive sterility and its application in aquatic breeding.
