奶白菜‘1170’核基因雄性不育系創(chuàng)制與利用

【摘要】:為了改進奶白菜核復(fù)等位基因遺傳的雄性不育系‘GMS3,園藝學(xué)性狀,以穩(wěn)定遺傳的奶白菜優(yōu)異自交系‘1170’為目標(biāo)品系,采用連續(xù)回交定向轉(zhuǎn)育方法選育新的雄性不育系;以轉(zhuǎn)育成的新不育系為親本,與自交系配制雜交組合,通過品種比較試驗,篩選優(yōu)良雜交種。主要研究結(jié)果如下:1.以核不育系‘GMS3’為測交親本,鑒定待轉(zhuǎn)育可育品系‘1170’的基因型,證明其在核不育位點上為純合顯性的恢復(fù)基因型,即MsfMsf。2.根據(jù)目標(biāo)品系的基因型(MsfMsf),設(shè)計定向轉(zhuǎn)育方案,將‘GMS3’的雄性不育基因轉(zhuǎn)入目標(biāo)品系‘1170’中,獲得了新的奶白菜核基因雄性不育系,以及其相應(yīng)的甲型“兩用系”和“臨時保持系”。3.轉(zhuǎn)育成的新不育系育性鑒定結(jié)果表明,其不育株率和不育度均達到了100%,不育性遺傳穩(wěn)定,不育系的植物學(xué)性狀接近轉(zhuǎn)育目標(biāo)品系‘1170’,達到了定向轉(zhuǎn)育的效果。4.以轉(zhuǎn)育成的不育系為母本,與10個自交系配制雜交組合,進行品種比較試驗,對各雜交組合的農(nóng)藝性狀進行鑒定。綜合各雜交組合產(chǎn)量、株形、幫色等調(diào)查結(jié)果,篩選出2個優(yōu)異雜交組合‘Z10’和‘Z34’。
[Abstract]:In order to improve the genoallelic male sterile line (GMS3) and horticultural character of Chinese cabbage (Brassica campestris L.), a new male sterile line was selected by continuous backcross directional transformation with the stable inheritance of the excellent inbred line of Chinese cabbage, Qiu 1170'as the target line. The new sterile lines were used as parents to prepare hybrid combinations with inbred lines. The main results were as follows: 1. Using GMS 3'as the test parent, we identified the genotypes of the fertile line, Y1170', which was to be transfered, and the results were as follows: (1) the genetic male sterile line (GMS 3') was used as the test parent to identify the genotypes of the fertile line, Y1170'. It was proved that the GMS gene was homozygous and dominant, that is, MsfMsf.2.According to the genotype of the target strain, MsfMsfGMS3 'gene was transferred into the target line, Y1170', according to the targeted genotype MsfMsfGMS3', and the result showed that the GMS3 gene was homozygous and dominant, I. E. MsfMsf.2. A new genic male sterile line of Chinese cabbage was obtained, and its corresponding A "dual purpose line" and "temporary maintainer line" .3.The results of fertility identification of the new sterile line showed that the sterile plant rate and sterility degree reached 100%, and the genetic stability of sterility was stable. The botanical characters of the male sterile line were close to the target line of transformation, and the effect of directional transfer was achieved. 4. Using the transformed sterile line as the female parent, the hybrid combination was prepared with 10 inbred lines, and the variety comparison experiment was carried out. The agronomic characters of each cross combination were identified. Based on the investigation results of yield, plant shape and band color of each cross combination, two excellent cross combinations, Z10 'and Z34', were screened out.
