石榴PgUFGT基因克隆及表達分析

【摘要】:利用RT-PCR和RACE方法,從石榴(Punica granatum L.)果皮中克隆到一個類黃酮糖基轉(zhuǎn)移酶(UFGT)基因(PgUFGT)全長cDNA序列(GenBank登錄號為KF841620)。PgUFGT基因編碼區(qū)1 476bp,編碼491個氨基酸。PgUFGT蛋白具有保守PSPG基序、UDP-糖基轉(zhuǎn)移酶家族結(jié)構(gòu)域和UDP-葡萄糖醛酸基/葡萄糖基轉(zhuǎn)移酶保守域(UDPGT),與其他植物UFGT蛋白一致性較高;系統(tǒng)進化樹分析結(jié)果表明,PgUFGT屬于類黃酮3-O-糖基轉(zhuǎn)移酶類。熒光定量qRT-PCR結(jié)果表明,PgUFGT基因在‘紅寶石’和‘水晶甜’2個石榴品種的發(fā)育期內(nèi)具有不同的表達模式,PgUFGT在‘紅寶石’石榴中有2個轉(zhuǎn)錄表達高峰,而在‘水晶甜’石榴中僅有1個表達高峰,表明PgUFGT可能在2個石榴品種中具有不同的催化作用。該研究結(jié)果為進一步研究石榴果實色澤形成的分子機制奠定了基礎。
[Abstract]:Using RT-PCR and RACE methods, A full-length cDNA sequence of flavonoid glycosyltransferase (granatum granatum) gene was cloned from the pericarp of Punica granatum L.). GenBank accession number is 1 476 BP, encoding 491 amino acids. PgUFGT protein has conserved PSPG motif and UDP- glycosyltransferase family knot. The conserved domain of UDP-glucuronyl / glucosyltransferase (UDP-glucuronyl / glucosyltransferase) conserved with UFGT protein in other plants. Phylogenetic tree analysis showed that PgUFGT belonged to flavonoid 3-Oglycosyltransferase. Fluorescence quantitative qRT-PCR showed that PgUFGT gene had different expression patterns during the development period of 'ruby' and 'crystal sweet' pomegranate. There were two peaks of transcription expression in 'ruby' pomegranate. However, there was only one peak expression in 'crystal sweet' pomegranate, indicating that PgUFGT may have different catalytic effect in two pomegranate varieties. The results laid a foundation for further study on the molecular mechanism of fruit color formation of pomegranate.
