水稻葉色突變體ygr的遺傳分析與基因定位

【摘要】:葉色突變體是研究水稻葉綠體發(fā)育、光合色素合成與降解的理想材料。前期研究在水稻保持系大面積繁殖時(shí)發(fā)現(xiàn)一株轉(zhuǎn)綠型淡黃葉突變體ygr,為了闡明其葉色調(diào)控機(jī)理,本試驗(yàn)比較了ygr與野生型的主要農(nóng)藝性狀和光合色素含量,并對(duì)該突變體進(jìn)行了遺傳分析和基因定位。結(jié)果表明,與野生型相比,ygr的始穗期推遲2 d,株高、單株有效穗數(shù)、穗長(zhǎng)、穗粒數(shù)、結(jié)實(shí)率和千粒重等主要農(nóng)藝性狀均無(wú)顯著差異,而苗期和抽穗期的葉綠素a、葉綠素b、總?cè)~綠素和類胡蘿卜素等光合色素含量均顯著降低。遺傳分析表明,ygr葉色突變性狀受單隱性核基因控制,暫時(shí)將其命名為YGR。以ygr與日本晴雜交的F_2群體作為定位群體,將YGR基因定位在水稻第1號(hào)染色體分子標(biāo)記WY8到WY20之間,其遺傳距離分別為0.2 cM和0.5 cM。本研究結(jié)果為該基因的克隆和功能分析以及解析其突變機(jī)制奠定了基礎(chǔ)。
[Abstract]:Leaf color mutants are ideal materials for chloroplast development, photosynthetic pigment synthesis and degradation in rice. In order to elucidate the mechanism of leaf color regulation, a green yellowish leaf mutants ygrr was found in large area reproduction of rice maintainers. The main agronomic characters and photosynthetic pigment contents of ygr and wild type were compared, and the genetic analysis and gene mapping of the mutant were carried out. The results showed that compared with the wild type, the beginning of ear was delayed by 2 days, the plant height and the number of effective panicles per plant were delayed. There were no significant differences in main agronomic characters such as ear length, grain number per ear, seed setting rate and 1000-grain weight, etc. However, chlorophyll a, chlorophyll b, total chlorophyll and carotenoid decreased significantly in seedling and heading stage. Genetic analysis showed that leaf color mutation traits of ygr were controlled by single recessive gene. The YGR gene was located between WY8 and WY20 on chromosome 1 of rice. The genetic distances were 0.2 cm and 0.5 cm, respectively. The results laid a foundation for cloning and functional analysis of the gene and for the analysis of its mutation mechanism.
